Angiotensin II Type 1 Receptor Tachyphylaxis Is Defined by Agonist Residence Time

Summary: This study investigated the phenomenon of tachyphylaxis in the angiotensin II type 1 receptor (AT(1)R) and found that ligand-binding kinetics play an important role in tachyphylaxis. The endogenous tachyphylactic agonist Ang II had a longer residence time at the receptor compared to analogs, resulting in sustained G(q) protein activation and recruitment of beta-arrestin. Furthermore, Ang II led to sustained receptor internalization, preventing further receptor responses.

Summary: This study used advanced RNA-sequencing techniques and bioinformatics analysis to identify gene signatures that can predict the responsiveness to pembrolizumab monotherapy. The T-cell-inflamed gene expression profile was associated with a better treatment response, while angiogenesis, monocytic myeloid-derived suppressor cell, and stroma/epithelial-mesenchymal transition/TGF-beta gene signatures were associated with a lower treatment response.

Summary: Despite modest clinical improvement with AVA therapy in ovarian cancer, adaptive resistance is a common problem and alternative options are limited. This study investigates the effectiveness of a GLS inhibitor (GLSi) in combination with AVA therapy, and found that it had significant cytotoxic effects and reduced tumor growth. Additionally, GLSi showed promising results in reducing metabolic conversion and had robust antitumor effects when administered after the emergence of resistance to AVA therapy.

Modulation of Slow Desensitization (Tachyphylaxis) of Acid-Sensing Ion Channel (ASIC)1a

Summary: In this study, the partial reversibility of ASIC1a's tachyphylaxis was demonstrated through long-term whole-cell recording of CHO cells transfected with rASIC1a cDNA, indicating that tachyphylaxis represents a slow desensitization of ASIC1a. Slow and steady-state desensitization were found to be independent processes, with certain drugs enhancing slow desensitization and others attenuating it. The effects of various conditions on ASIC1a response amplitude were correlated with the development of slow desensitization.

Global burden of paediatric vision impairment: a trend analysis from 1990 to 2017

Summary: The global prevalence of distance and/or near vision impairment for children aged 1-14 in 2017 was 2.8%. The highest DALYs were observed in teenagers, with a positive correlation between DALY rate of refractive disorders and national socioeconomic development. From 1990 to 2015, DALY numbers remained stable but rates showed a significant trend of reduction in all paediatric age groups.

Unplanned reoperation following vitreoretinal surgery

Summary: The purpose of this study was to evaluate the incidence and reasons of unplanned reoperations following vitreoretinal surgery. It was found that unplanned reoperations were not very common and mostly occurred in patients with proliferative vitreoretinopathy, rhegmatogenous retinal detachment, and tractional retinal detachment. Lower preoperative best-corrected visual acuity may indicate an increased chance of future unplanned reoperations.

The study found that retinotomy with/without retinectomy is effective in the majority of patients with RRD associated with advanced PVR, but additional surgeries are needed in a significant number of eyes to achieve final anatomical success.

Optic pit-like maculopathy in a patient without any detectable disc anomalies

Summary: Macular schisis refers to the splitting of inner retinal layers at the macula, which can be associated with various conditions. Cases of retinoschisis without optic disc pit have been reported, suggesting possible connections between macular neuroretinal tissue and the vitreous cavity. Further studies are needed to explore the mechanisms of this condition with more subjects.
