Aberrant promoter CpG islands methylation of tumor suppressor genes in cholangiocarcinoma

Aberrant DNA methylation of 5'-CpG islands located within gene promoters has been identified as a mechanism for transcriptional inactivation of tumor suppressor genes. To ascertain the mechanism of gene promoter hypermethylation in cholangiocarcinoma (CC), we investigated promoter methylation status of the candidate genes ID4, DLC-1, and SFRP1 in 41 CCs, 19 adjacent nontumor tissues, and 15 normal liver tissues using methylation-specific PCR (MSP). The frequencies of DNA methylation were: 57.5% (23 of 40) for ID4, 14.3% (5 of 35) for DLC-1, and 63.4% (26 of 41) for SFRP1, respectively. In contrast, a low frequency of methylation was detected in nontumor tissues. In addition, hypermethylated status of these genes was detected in three kinds of CC cell lines. Moreover, the downregulated expression of these genes in these cells was restored by treatment with 5-aza-2'-deoxycytidine, a DNA methyltransferase inhibitor. Taken together, these results suggest that abet-rant DNA methylation may contribute to the tumorigenesis of cholangiocarcinoma.

Summary: Recent studies have shown that 2D germanium telluride nanosheets coated with polyvinylpyrrolidone (PVP) have potential applications in nanomedicine for multispectral optoacoustic tomography and fluorescence imaging and therapy of inflammatory bowel disease. These nanosheets exhibit broad optical absorption in visible and near-infrared ranges, providing deeper tissue images than other materials. Additionally, when orally administered, they show therapeutic efficacy similar to or higher than traditional medication for inflammatory diseases in the gastrointestinal tract.

Molecular and radiopathologic spectrum between HCC and intrahepatic cholangiocarcinoma

Summary: In this study, we performed an integrative analysis of transcriptome profiles associated with radiopathologic features to identify four subtypes of primary liver cancers (LCs). Our findings revealed intermediate subtypes between hepatocellular carcinoma (HCC) and intrahepatic cholangiocarcinoma (iCCA), and provided insights into their molecular spectrum and progression mechanisms.

Noninvasive surrogates are poor predictors of liver fibrosis in patients with Fontan circulation

Summary: Patients with Fontan circulation have a high incidence of liver fibrosis and cirrhosis. This study found that transient elastography (TE) and the enhanced liver fibrosis (ELF) test were unable to predict the degree of liver fibrosis in Fontan patients. Elevated N-terminal pro-brain natriuretic peptide levels and ventricular end-diastolic pressure may be associated with advanced liver fibrosis.

MRI features of histologic subtypes of hepatocellular carcinoma: correlation with histologic, genetic, and molecular biologic classification

Summary: HCC is a diverse group of tumors in terms of histology, genetics, and protein expression. Advancements in imaging techniques have made imaging diagnosis a critical part of managing HCC in the clinical setting. There is increasing evidence of correlation between imaging phenotypes and histologic, molecular, and genetic characteristics of different HCC subtypes. Further research on radiological characteristics of HCC subtypes may enable non-invasive diagnosis and serve as biomarkers for prognosis, molecular characteristics, and therapeutic response.

Up-to-Date Pathologic Classification and Molecular Characteristics of Intrahepatic Cholangiocarcinoma

Summary: Intrahepatic cholangiocarcinoma (iCCA) is an aggressive primary liver malignancy with distinct histopathologic features and molecular profiles. The classification of small duct type and large duct type iCCA provides important insights into the tumor behavior and prognosis. Understanding the tumor microenvironment and genomic characteristics can help develop precision therapeutics for patients with iCCA.

Albumin, filamin-A and cytokeratin 19 help distinguish intrahepatic cholangiocarcinoma from extrahepatic adenocarcinoma

Summary: In this study, a method was developed to differentiate intrahepatic cholangiocarcinomas (iCCAs) from adenocarcinomas originating from extrahepatic organs adjacent to the liver. The combination of albumin mRNA in situ hybridization (ISH) and immunohistochemistry (IHC) staining for filamin-A and cytokeratin 19 (CK19) showed better diagnostic performance than albumin ISH alone. The newly developed iCCA-score consisting of these three markers can accurately differentiate iCCA from extrahepatic adenocarcinomas.

Summary: This study evaluated the clinical activity of FOLFOX plus anti-HER2 antibody trastuzumab as a second-line or third-line treatment for HER2-positive biliary tract cancer. The overall response rate was 29.4% and the disease control rate was 79.4%. The treatment showed promising activity with acceptable toxicity in HER2-positive biliary tract cancer.

SEPT9 Expression in Hepatic Nodules: An Immunohistochemical Study of Hepatocellular Neoplasm and Metastasis

Summary: The methylated SEPT9 DNA (mSEPT9) in plasma is an FDA-approved screening biomarker in colorectal cancer and is emerging as a promising diagnostic and prognostic biomarker in hepatocellular carcinoma (HCC). In this study, the SEPT9 protein expression in various hepatic tumors was evaluated using immunohistochemistry (IHC), and its correlation with demographics, risk factors, tumor characteristics, and oncologic outcomes was analyzed. The findings suggest that SEPT9 may be implicated in liver carcinogenesis and that SEPT9 staining by IHC could serve as an adjunct diagnostic biomarker with potential prognostic ramifications.

Summary: This study aimed to develop and validate a simple scoring system to predict 2-year recurrence after surgical resection (SR) for hepatocellular carcinoma (HCC). By studying 2359 HCC patients who underwent SR in 17 centers, variables associated with 2-year recurrence were identified and used to create the Early Recurrence Score (ERS). The ERS is a user-friendly staging system that can help design personalized surveillance strategies and adjuvant therapy trials.
