Apoptotic markers in a prostate cancer cell line: Effect of ellagic acid

Ellagic acid (EA) inhibits cell growth and induces apoptosis in cultured cells; however, the precise molecular mechanism involved in EA-induced apoptosis in prostate cancer cells is unknown. The aim of the present study was to delineate possible apoptotic pathway(s) involved in the EA-mediated chemotherapeutic effects in the LNCaP human prostatic cancer cell line. EA produced anti-proliferative effects through inhibition of rapamycin (mTOR) activation and a reduction in intracellular levels of -catenin. Moreover, we demonstrated that EA induced apoptosis via downregulation of the anti-apoptotic proteins, silent information regulator 1 (SIRT1), human antigen R (HuR) and heme oxygenase-1 (HO-1). EA modulated the expression of apoptosis-inducing factor (AIF) resulting in a significant increase in reactive oxygen species (ROS) levels and the activation of caspase-3. Finally, we demonstrated that EA reduced both transforming growth factor- (TGF-) and interleukin-6 (IL-6) levels. EA treatment resulted in the increased expression of the tumor suppressor protein p21 and increased the percentage of apoptotic cells. In conclusion, the results suggest that EA treatment represents a new and highly effective strategy in reducing prostate cancer carcinogenesis.

Chemical, Antioxidant and Biological Studies of Brassica incana subsp. raimondoi (Brassicaceae) Leaf Extract

Summary: Brassica incana subsp. raimondoi is an endemic taxon found in eastern Sicily, growing on steep limestone cliffs. This research investigated the phytochemical profile, antioxidant properties, cytotoxicity, and Artemia salina toxicity of its leaf extract. The extract contained various phenolic acids, flavonoids, and carotenoids, and showed different antioxidant properties. It was non-toxic to normal cells, but exhibited cytotoxic effects on colon cancer and hepatoma cells. It also protected against oxidative stress caused by hydrogen peroxide. These findings suggest that B. incana subsp. raimondoi could be a potential source of bioactive molecules.

Ethyl Protocatechuate Encapsulation in Solid Lipid Nanoparticles: Assessment of Pharmacotechnical Parameters and Preliminary In Vitro Evaluation for Colorectal Cancer Treatment

Summary: Colorectal cancer is a common tumor disease, and the use of naturally derived products for its treatment can be beneficial. The research focused on the encapsulation of ethyl protocatechuate (PCAEE) in solid lipid nanoparticles (SLN) to assess its effectiveness. SLN prepared with Tween 80 showed promising physicochemical properties and encapsulation efficiency, effectively penetrated and released PCAEE, and exhibited greater cytotoxicity on tumor cells. Therefore, further studies on this formulation for colorectal cancer treatment are suggested.

Near-Infrared-Responsive Choline-Calix[4]arene-Gold Nanostructures for Potential Photothermal Cancer Treatment

Summary: This study presents a novel method to prepare water-dispersible gold nanostructures with near-infrared localized surface plasmon resonance (LSPR) properties. These nanostructures were synthesized using NADH as a reducing agent and an amphiphilic choline-calix[4]arene derivative (CholCalix) forming micelles as a template. Characterization of the CholCalix-Au nanostructures was performed using UV-visible spectrophotometry, Raman spectroscopy, and atomic force microscopy. The agglomeration of the nanostructures due to multiple crosslinking interactions was observed and supported by modeling simulation. The CholCalix-AuNPs demonstrated effective anticancer photothermal-induced effect on human breast cancer cells irradiated with biofriendly light at 808 nm.

Standardized Extract from Wastes of Edible Flowers and Snail Mucus Ameliorate Ultraviolet B-Induced Damage in Keratinocytes

Summary: Several studies have found that snail mucus has emollient, regenerative, and protective properties for maintaining healthy skin conditions. The mucus from Helix aspersa muller has shown beneficial properties like antimicrobial activity and wound repair capacity. By enriching snail mucus with antioxidant compounds derived from edible flower waste, researchers have demonstrated that the flower waste extract boosts the antioxidant activity of snail mucus, providing cytoprotective effects against UVB radiation in keratinocytes.

Summary: Our research team conducted a study on Brassica fruticulosa subsp. fruticulosa, an edible plant used for treating various ailments. We found that the leaf extract exhibited good antioxidant properties, with the secondary compounds being more potent than the primary ones. Further investigation revealed that the phenolic compounds in the extract possessed antioxidant properties and inhibited the proliferation of human colorectal epithelial adenocarcinoma cells.

Summary: The aim of this study was to investigate the capability of MGN-NLC as a potential drug delivery system for ocular administration. The in vitro and ex vivo results showed that MGN-NLC exhibited no cytotoxic effects, maintained the antioxidant role of MGN, and could permeate and accumulate in ocular tissues. These findings suggest a potential application of MGN-NLC in the treatment of oxidative stress-related ocular diseases.
