Natural killer-like T CD3+/CD16+CD56+ cells in chronic lymphocytic leukemia: Intracellular cytokine expression and relationship with clinical outcome

The clinical significance of NK-like T CD3(+)/CD16(+)CD56(+) cells in chronic lymphocytic leukemia (CLL) is still a subject of controversy. There are few previous descriptions that this cell population can be qualitatively or quantitatively deficient in CLL patients. In the present study we investigated the clinical value of CD3(+)/CD16(+)CD56(+) cells as predictors of disease progression. We assessed the frequencies of CD3(+)/CD16(+)CD56(+) cells by the flow cytometry in a group of 300 CLL patients. The percentage of CD3(+)/CD16(+)CD56(+) cell population expressed as the percentage of CD3(+) lymphocyte compartment showed an inverse correlation with ZAP-70 and CD38. Additionally, the CD3(+)/CD16(+)CD56(+) showed an inverse correlation with LPL/ADAM29 ratio. Likewise, the ability of these cells to cytokine expression correlated with ZAP-70 expression. A positive correlation between percentage CD3(+)/CD16(+)CD56(+) cells and TFS was found. The decreased percentage of these cells was associated with higher death risk in CLL patients. Furthermore, the percentage of CD3(+)/CD16(+)CD56(+) cells was significantly decreased in patients who showed progression of disease. This study suggests that assessment of CD3(+)/CD16(+)CD56(+) cells may be helpful in determining a worsening of clinical course. Monitoring of these cell numbers and function may provide useful information for determining disease activity. Especially, it could be intormative to look at these cells in patients with stage 0 CLL. For this patient group immunological control and dysfunction are probably important factors.

Summary: PAH can be caused by disorganized endothelial cell proliferation in the pulmonary arterioles, and factors such as genetic susceptibility, hypoxia, inflammation, drug response, and toxins may contribute to its development. The interaction of CD200 and CD200R in immune response modulation plays a crucial role in vascular remodeling associated with PAH. The dysregulation of CD200 and CD200R expression may be involved in the pathogenesis of various immune diseases, making these molecules potential markers for distinguishing different types of PAH.

Reduced Percentage of CD14dimCD16+SLAN+ Monocytes Producing TNF and IL-12 as an Immunological Sign of CLL Progression

Summary: Monocytes, particularly non-classical subpopulations, may play an important role in chronic lymphocytic leukemia (CLL) pathogenesis. This study assessed monocyte subsets divided based on SLAN and CD16 expression in CLL patients. The findings suggest an increased percentage of non-classical monocytes producing TNF and IL-12, while intermediate monocytes showed a higher percentage of cells synthesizing anti-inflammatory IL-10. The percentage of CD14(dim)CD16(+)SLAN(+) monocytes producing TNF and IL-12 decreased with CLL stage and correlated with prognostic factors. Notably, CLL patients requiring treatment had a lower percentage of CD14(dim)CD16(+)SLAN(+) monocytes producing TNF and IL-12, potentially indicating a beneficial effect of non-classical monocytes on anti-tumor response.

Programmed Cell Death Protein-1 Upregulation in Response to SARS-CoV-2 in Juvenile Idiopathic Arthritis: A Case-Control Study

Summary: This study aimed to evaluate the impact of COVID-19 disease on patients with childhood rheumatic diseases. The study found significantly higher levels of SARS-CoV-2 antibodies in JIA patients compared to the control group, and these levels were correlated with the serological response to SARS-CoV-2. Additionally, serum PD-1 levels were also significantly higher in JIA patients and control volunteers who were seropositive for SARS-CoV-2 antibodies. The findings highlight the need for further observation of these detected abnormalities.

Prognostic Potential of Galectin-9 mRNA Expression in Chronic Lymphocytic Leukemia

Summary: This study explores the potential role of Galectin-9 (Gal-9) as a novel prognostic biomarker in chronic lymphocytic leukemia (CLL). Through quantification of Gal-9 mRNA expression in purified B-lymphocytes of 100 CLL patients, the study reveals the upregulation of Gal-9 mRNA in malignant B-cells, which is closely associated with unfavorable prognostic markers and a shorter time to treatment. Furthermore, the study suggests that EBV coinfection may worsen prognosis in CLL by inducing upregulation of Gal-9 expression.
