Summary: The role of circ_0002019 in gastric cancer (GC) was investigated in this study. It was found that circ_0002019 was highly expressed in GC tissues and cells. Silencing circ_0002019 suppressed the proliferation, migration, and invasion of GC cells. Mechanistically, circ_0002019 activated the NF-κB pathway by increasing the stability of TNFAIP6 mRNA through PTBP1. These findings indicate that circ_0002019 plays a critical regulatory role in the progression of GC.

Summary: Our study revealed that INHBB is significantly upregulated in gastric cancer (GC) and its increased expression is associated with poor survival. INHBB promotes the proliferation, migration, and invasion of GC cells in vitro. Additionally, activin B reprograms fibroblasts into cancer-associated fibroblasts (CAFs) and promotes GC development. The high expression of INHBB in GC cells activates the NF-kappa B pathway in normal gastric fibroblasts via the secretion of activin B, leading to fibroblast proliferation, migration, and invasion. Activin B induces NF-kappa B pathway activation in fibroblasts through controlling TRAF6 autoubiquitination and TAK1 phosphorylation. Activated fibroblasts release pro-inflammatory factors IL-1 beta, which induce p65 phosphorylation in GC cells. p65 directly binds to the INHBB promoter, increasing INHBB transcription in GC cells and establishing a positive regulatory feedback loop that promotes GC progression.

Summary: The role of tumor-associated macrophages (TAMs) and the regulatory mechanisms of macrophage activation in prostate cancer (PCa) are poorly understood. In this study, it was found that PCa growth in mice with macrophage-specific Ubc9 deficiency was suppressed due to enhanced CD8+ T cell response. The study also identified STAT4 as a crucial UBC9-mediated SUMOylation target, with K350 as the major modification site. Furthermore, inhibition of UBC9 with the inhibitor 2-D08 increased the antitumor effect of TAMs and expression of PD-1 on CD8+ T cells, providing valuable insights for cancer therapy.
