Summary: Juglone and KPT6566 inhibit the growth and colony formation of Caco-2 cells and induce apoptosis. They also downregulate the expression of cell cycle-related proteins, leading to suppression of Caco-2 cell proliferation and colony formation. The experiments show that Juglone or KPT6566 can kill both tumor-initiating and non-tumor-initiating Caco-2 cells with similar efficacy.

Summary: In addition to their protective roles in hemostasis and innate immunity, platelets have been found to play diverse and adverse roles in the development, progression, and outcome of various human malignancies. Elevated circulating platelet counts have been consistently associated with the onset and prognosis of different cancers, particularly lung cancer. This review focuses on the association of lung cancer with circulating platelet count, platelet-mediated immunosuppression, platelet-derived microparticles, cancer-related platelet activation and thrombosis, and the potential use of antiplatelet agents in cancer therapy.

Summary: In this study, A549 human lung carcinoma cells were cultured long-term on micropillar-embossed dishes to investigate the influence on the pathological state of the cells. Results showed that the cells exhibited more aggressive characteristics, along with morphological deformation in nuclei and cell-cycle arrest, leading to suppressed proliferation. This study highlights the potential use of pillar-based mechanical stimuli as an in vitro cell culture strategy to induce more aggressive tumorigenic cancer cell models.

Summary: The Integrated Stress Response (ISR) plays a crucial role in the development of lung adenocarcinoma (LUAD), particularly in KRAS-driven lung tumorigenesis. Inhibition of ISR may provide a potential therapeutic option, significantly reducing tumor growth and prolonging survival.

Summary: The lung immune prognostic index (LIPI) is proposed as a tool to differentiate prognosis and treatment benefit from immune checkpoint inhibitors in advanced non-small cell lung cancer. A study found that LIPI can identify subgroups with significantly different survival following combination therapies including ICIs.

Summary: The study aims to assess the applicability of TiY in theranostics, from in vivo visualization to therapeutic effect towards tumor-initiating cells (TIC) in cancer mouse models. Through cell experiments and animal model studies, we demonstrated the selective identification and therapeutic effect of TiY on TIC in animal models.

Summary: Despite improvements in therapies and screening strategies, lung cancer prognosis remains poor, especially for patients with metastatic tumors. Cancer stem cells (CSCs) are a major cause of the low survival rate, and microRNA miR-486-5p has been identified as a potential therapeutic target for lung cancer.

Summary: Extracellular vesicles (EVs) released from non-small cell lung cancer (NSCLC) cells play a crucial role in promoting cancer progression. There are differences in the secretion profile and the ability to induce phenotypic changes in non-tumorigenic cells among EVs derived from different NSCLC cell lines. EV-encapsulated RNA (EV-RNA) is identified as a key factor in mediating these phenotypes. The aggressiveness of NSCLC subtypes may be correlated with the ability of their respective EVs to induce cancerous phenotypes.

Summary: Extensive-stage small-cell lung cancer is a challenging disease to treat, and despite improvements in first-line therapy, resistance remains a significant issue. Recent progress in understanding the molecular biology of SCLC holds potential for molecularly informed therapeutic strategies for patients with SCLC.

Summary: In this study, the mechanism of miRNA-mediated transcriptional regulation in ZNF322A-driven oncogenic events was explored. miR-326 inhibited lung cancer progression by downregulating the activity and expression of ZNF322A, and its expression was negatively correlated with ZNF322A. The deregulated expression of miR-326 hyperactivated ZNF322A-driven oncogenic signaling, highlighting the potential of targeting the miR-326/ZNF322A axis as a therapeutic strategy for lung cancer patients.

Summary: The past decade has seen significant progress in the treatment of metastatic non-small-cell lung cancer, with the development of specific antibodies leading to improved survival for many patients. Currently, nearly all mNSCLC patients receive PD-1 or PD-L1 therapy in the first-line setting, except those with targetable oncogenes.

Summary: This study used Mendelian randomization analysis to investigate the causal association between the cathepsin family and lung cancer. The results suggest that elevated levels of cathepsin H increase the overall risk of lung cancer, adenocarcinoma, and lung cancer among smokers. Conversely, squamous carcinoma may lead to increased levels of cathepsin B. These findings have implications for the diagnosis and treatment of lung cancer.
