Cross-validation and refinement of the Stoffenmanager as a first tier exposure assessment tool for REACH

Objectives For regulatory risk assessment under REACH a tiered approach is proposed in which the first tier models should provide a conservative exposure estimate that can discriminate between scenarios which are of concern and those which are not. The Stoffenmanager is mentioned as a first tier approach in the REACH guidance. In an attempt to investigate the validity of the Stoffenmanager algorithms, a cross-validation study was performed. Methods Exposure estimates using the Stoffenmanager algorithms were compared with exposure measurement results (n=254). Correlations between observed and predicted exposures, bias and precision were calculated. Stratified analyses were performed for the scenarios handling of powders and granules (n=82), handling solids resulting in comminuting (n=60), handling of low-volatile liquids (n=40) and handling of volatile liquids (n=72). Results The relative bias of the four algorithms ranged between -9% and -77% with a precision of approximately 1.7. The 90th percentile estimate of one out of four algorithms was not conservative enough. Based on these statistics and analyses of residual plots the underlying algorithm was adapted. Subsequently, the calibration and the cross-validation dataset were merged into one dataset (n=952) used for calibrating the adapted Stoffenmanager algorithms. This new calibration resulted in new exposure algorithms for the four scenarios. Conclusions The Stoffenmanager is capable of discriminating among exposure levels mainly between scenarios in different companies. The 90th percentile estimates of the Stoffenmanager are verified to be sufficiently conservative. Therefore, the Stoffenmanager could be a useful tier 1 exposure assessment tool for REACH.

Recent pesticide exposure affects sleep: A cross-sectional study among smallholder farmers in Uganda

Summary: The study revealed an increased risk of sleep problems among smallholder farmers in Uganda in a pesticide exposure-dependent manner, particularly higher risk for female pesticide applicators. Further gender-stratified and longitudinal investigations are needed to confirm these findings.

Evaluation of two-year recall of self-reported pesticide exposure among Ugandan smallholder farmers

Summary: The study in Uganda evaluated smallholder farmers' recall of pesticide use and exposure determinants over a two-year period. Better recall was found in some aspects such as the use of active ingredients and PPE items. However, recall of major crops was poor, and a trend was observed where more individuals reported using active ingredients while fewer reported using PPE items after two years.

Summary: Assessment of occupational pesticide exposure in epidemiological studies of chronic diseases is challenging. This study found that different exposure assessment methods did not significantly affect risk estimates for prostate cancer, non-Hodgkin's lymphoma, and Parkinson's disease. Study design, publication year, and geographic location should be considered when evaluating the chronic health effects of occupational pesticide exposure.

Summary: Limited evidence exists regarding the exposure-effect relationship between lung-cancer risk and hexavalent chromium (Cr(VI)) or nickel. This study found that occupational exposure to Cr(VI) and nickel was associated with lung-cancer risks, particularly in men. In men, the highest quartile of cumulative exposure to CR(VI) and nickel were both significantly associated with increased odds ratios for lung cancer.

Temporal Trends in Variability of Respirable Dust and Respirable Quartz Concentrations in the European Industrial Minerals Sector

Summary: This study examines long-term trends in the variability of occupational exposure, specifically respirable dust and respirable quartz concentrations within the European industrial minerals sector. The European Industrial Minerals Association's Dust Monitoring Program (IMA-DMP) collected approximately 40,000 personal full-shift measurements since 2000. The study found that day-to-day variability played a major role, with within-worker variability higher for respirable quartz than for respirable dust. Meanwhile, between-worker variability decreased over time, but within-worker variability showed downward and upward temporal trends for both respirable dust and respirable quartz concentrations.

Development of a quantitative North and Central European job exposure matrix for wood dust

Summary: The study aimed to develop a multinational Job Exposure Matrix (JEM) for wood dust exposure in Northern and Central European countries. The results showed an annual decline of approximately 8% in wood dust exposure and significant differences in exposure levels between countries, with the highest levels in the United Kingdom. The jobs with the highest predicted exposure were floor layers and tile setters, wood-products machine operators, and building construction labourers.
