Summary: This study presents a surgical approach to induce demyelination in the rat sciatic nerve, followed by remyelination after Teriflunomide (TF) treatment. Histology and immunostaining reveal demyelination or myelin loss after sciatic nerve injury, with no self-recovery. The rotarod test confirms the motor function loss in nerve-damaged rats. Transmission electron microscopy shows axonal atrophy and inter-axonal gaps in these rats. TF administration restores motor function, repairs axonal atrophy with inter-axonal spaces, and promotes myelin secretion or remyelination. These findings demonstrate the potential of the surgical procedure and TF treatment in inducing and repairing demyelination.
