SCIATIC NERVE REGENERATION IS NOT INHIBITED BY ANTI-NGF ANTIBODY TREATMENT IN THE ADULT RAT

Elevated nerve growth factor (NGF) is believed to play a role in many types of pain. An NGF-blocking antibody (muMab 911) has been shown to reduce pain and hyperalgesia in pain models, suggesting a novel therapeutic approach for pain management. Since NGF also plays important roles in peripheral nervous system development and sensory nerve outgrowth, we asked whether anti-NGF antibodies would adversely impact peripheral nerve regeneration. Adult rats underwent a unilateral sciatic nerve crush to transect axons and were subcutaneously dosed weekly for 8 weeks with muMab 911 or vehicle beginning 1 day prior to injury. Plasma levels of muMab 911 were assessed from blood samples and foot print analysis was used to assess functional recovery. At 8-weeks post-nerve injury, sciatic nerves were prepared for light and electron microscopy. In a separate group, Fluro-Gold was injected subcutaneously at the ankle prior to perfusion, and counts and sizes of retrogradely labeled and unlabeled dorsal root ganglion neurons were obtained. There was no difference in the time course of gait recovery in antibody-treated and vehicle-treated animals. The number of myelinated and nonmyelinated axons was the same in the muMab 911-treated crushed nerves and intact nerves, consistent with observed complete recovery. Treatment with muMab 911 did however result in a small decrease in average cell body size on both the intact and injured sides. These results indicate that muMab 911 did not impair functional recovery or nerve regeneration after nerve injury in adult rats. Published by Elsevier Ltd. on behalf of IBRO.

Automated monitoring of respiratory rate as a novel humane endpoint: A refinement in mouse metastatic lung cancer models

Summary: This study aimed to identify objective non-invasive criteria for predicting imminent distress and mortality in metastatic lung tumor-bearing mice. The findings suggest that increases in respiratory rate can serve as an objective benchmark to signal the need for increased clinical observations or euthanasia. Adoption of this novel humane endpoint criterion would allow investigators time to collect tissue samples prior to spontaneous morbidity or death and significantly reduce the distress of mice in the terminal stages of these metastatic lung tumor models.

Summary: A new in- and ex vivo hybrid method for measuring GFR using the in vivo imaging system (IVIS) was developed as an alternative to the traditional filtration marker method with fluorescence molecular tomography (FMT). The results showed that the IVIS imaging measurement of inulin clearance in untreated, vehicle-treated and cyclosporine A-treated mice yielded comparable GFR values to those obtained with FMT imaging, demonstrating the feasibility and reliability of this alternative technique for assessing renal function in pharmacological and toxicological studies.

Introduction of gloved hand to cage induces 22-kHz ultrasonic vocalizations in male albino rats

Summary: This study found that the introduction of a gloved hand into a cage can trigger adult male albino rats to emit 22-kHz ultrasonic vocalizations. However, no 22-kHz vocalizations were found in female rats tested. Visual function, serum corticosterone, and testosterone levels did not show significant differences between responders and non-responders.

Summary: This study investigated the protective effects and mechanisms of human placenta derived mesenchymal stem cells (hPMSCs) transplantation in a rat model of hypoxic-ischemic encephalopathy (HIE). The results showed that hPMSCs transplantation reduced apoptosis and improved long-term neurological prognosis. Furthermore, the downregulation of Sema 3A/NRP-1 expression and activation of the PI3K/Akt/mTOR signaling pathway played a key role in the protective effects of hPMSCs.

Probing the Neurophysiology of Temporal Sensitivity in the Somatosensory System Using the Mismatch Negativity (MMN) Sensory Memory Paradigm

Summary: This study evaluated electrophysiological discrimination of parametric somatosensory stimuli in healthy young adults to understand how the brain processes the duration of tactile information. The results showed that participants did not electrophysiologically discriminate between 100 and 115 ms, but they exhibited distinct electrophysiological responses when the deviant stimuli were 130, 145, and 160 ms. These findings contribute to a better understanding of tactile sensitivity in different clinical conditions.

Summary: The basolateral amygdaloid complex (BLA) is closely involved in the formation of emotional memories, including both aversive memory and contextual fear memory. Acute sleep deprivation (SD) disrupts the acquisition of tone-associated fear memory in juvenile rats, but has no significant effect on contextual fear memory. Slow network oscillation in the amygdala contributes to the formation of amygdala-dependent fear memory in relation to sleep.
