Summary: v-Src alters the cytotoxicities of microtubule-targeting agents (MTAs) and polo-like kinase 1 (PLK1) inhibitor, but not DNA-damaging anticancer drugs. It induces mitotic slippage of MTAs-treated cells, leading to the generation of proliferating tetraploid cells. Treatment with Aurora kinase inhibitor strongly induces cell death in cells expressing v-Src.

Summary: This Policy Review compares the participation rates of China with the USA, EU, Japan, and South Korea in clinical trials for innovative anticancer drugs. The results show that China's participation rates are significantly lower than those of the other countries in both overall and synchronous participation. Despite an increase in participation over the past decade, China still lags behind the other regions, particularly in terms of synchronous participation and early participation rates.

Summary: Nigericin shows promising anticancer effects in human osteosarcoma by inhibiting tumor cell proliferation, inducing apoptosis, and targeting the SRC/STAT3 signaling pathway. The drug also inhibits tumor migration and invasion, and shows potential for clinical application in osteosarcoma treatment.

Summary: Drug repositioning is an effective approach in drug discovery, and this study evaluated the modulation of p8 subunit activity in TFIIH complex by several drugs. Out of 60 drugs tested, 7 showed potential interactions with p8 subunit and thermal destabilization. Further investigation is needed to explore their potential as anticancer drugs.

Summary: This review presents and critically discusses the chemistry and biosynthesis of allicin, its pharmacokinetic profile, its anticancer mechanisms and molecular targets, and its selectivity towards tumor cells. The research carried out so far reveals that allicin suppresses the growth of different types of tumors by targeting multiple signaling pathways associated with cancer development. Future research directions are outlined to further explore the potential of allicin as a natural product for cancer treatment.

Summary: Polymersomes are biomimetic cell membrane-like model structures that have gained attention for their biocompatibility, versatility, and tunable properties. They can encapsulate cargoes and be triggered to disassemble in response to stimuli, making them suitable for photodynamic therapy. This review explores the current advances and applications of light-responsive polymersomes in anticancer therapy.

Summary: This study analyzed the recent research on polyoxometalates as anticancer agents, focusing on their effects on the cell cycle. The findings showed that polyoxometalates can have diverse effects on the cell cycle, including protein expression, mitochondrial effects, ROS production, cell death, and cell viability. Comparison with clinically approved drugs revealed better results for polyoxometalates in many cases, suggesting their potential as alternative cancer therapies.

Summary: The pyrimidine and fused pyrimidine ring systems are important in inhibiting the c-Src kinase. Despite its association with cancer, the exploration of Src kinase by medicinal chemists is still limited, making it a niche pathway. This review focuses on the activation process of Src kinase, the chemistry of pyrimidine ring and its synthetic routes, as well as recent developments in c-Src kinase inhibitors containing pyrimidine and their biological activity, structure-activity relationship (SAR), and selectivity.

Summary: Anticancer drugs are highly hazardous due to their genotoxic, carcinogenic and teratogenic mechanisms of action. The hydrolytic stability of commonly used anticancer drugs was investigated, with results showing that cyclophosphamide and ifosfamide were unstable while other compounds displayed high stability. Further experiments revealed that cyclophosphamide and ifosfamide would be quite persistent in terms of hydrolytic stability at environmentally relevant temperatures.
