Summary: In this study, the production of recombinant human transforming growth factor-beta1 (rhTGF-beta 1) in Chinese hamster ovary (CHO) cells was enhanced through characterization and optimization. It was found that mature rhTGF-beta 1 was mostly produced in the form of latent rhTGF-beta 1 in CHO-rhTGF-beta 1 cells, and a significant amount of unprocessed precursors was also produced. Overexpression of PACEsol in CHO-rhTGF-beta 1 cells effectively cleaved the unprocessed precursors, resulting in increased production of mature rhTGF-beta 1.

Summary: Tau protein is involved in Alzheimer's disease due to its detachment from microtubules and formation of insoluble intracellular aggregates. Recent studies indicate the expulsion of Tau seeds in the extracellular domain and their prion-like spread between neurons. The cytokine Transforming Growth Factor-beta 1 (TGF-beta 1) plays a role in immunomodulation and neuroprotection in the brain and is also involved in cytoskeletal reorganization. Targeting extracellular Tau using TGF-beta-triggered microglia-mediated approach could be a strategy to combat Alzheimer's disease.

Summary: Primary myelofibrosis (PMF) is a disease characterized by abnormal cytokine production, leading to bone marrow fibrosis. This study found that the transmembrane protein GARP plays a role in activating TGF-beta 1, a profibrotic cytokine, in PMF. Blocking the activation of TGF-beta 1 by GARP-expressing cells reduced fibrosis and clonal expansion of transformed cells. The therapeutic effects were attributed to targeting GARP on regulatory T lymphocytes (Tregs) and increasing CD8 T-cell-mediated immune reaction.

Summary: This study developed fibrosis-mimicking models using 3D co-culture devices to evaluate the effects of TGF-beta and TGF-beta inhibitor treatment on renal fibrosis. The expression of fibrosis marker in cells increased upon TGF-beta treatment, while vessel density in the vascular compartment varied with TGF-beta and TGF-beta inhibitor treatment. Multiplex ELISA analysis showed increased levels of inflammatory cytokines and growth factors in the TGF-beta-stimulating 3D models, mimicking the fibrotic microenvironments of human kidneys. This study may lead to the construction of a human renal fibrosis-mimicking device model beyond traditional culture experiments.
