Taxol®-induced phosphatidylserine exposure and microvesicle formation in red blood cells is mediated by its vehicle Cremophor® EL

The conventional clinical formulation of paclitaxel (PTX), Taxol (R), consists of Cremophor (R) EL (CrEL) and ethanol. CrEL-formulated PTX is associated with acute hypersensitivity reactions, anemia and cardiovascular events. In this study, the authors investigated the effects of CrEL-PTX on red blood cells (RBCs) and compared these with the effects observed after exposure to the novel nanoparticle albumin-bound PTX, marketed as Abraxane r. Results: The authors demonstrate that CrEL is primarily responsible for RBC lysis and induction of phosphatidylserine exposure. Phosphatidylserine-exposing RBCs showed increased association with endothelial cells in culture. The authors also identified CrEL as being responsible for vesiculation of RBCs. This is the first time that excipients have been shown to be involved in microvesicle formation. Microvesicles were taken up by endothelial cells. Conclusion: These results offer new insights into the side effect profile of Taxol, which is likely to have implications for patients with erythrocyte disorders. Abraxane did not induce any of these effects on RBCs, indicating that the choice of excipients can have a pronounced influence on the efficacy and side effects of drug molecules.

The results showed no significant relationship between patient satisfaction with information and adherence, despite the high level of satisfaction with the information. Membrane curvature and PS localize coagulation proteins to filopodia and retraction fibers of endothelial cells

Summary: Prior reports have indicated that convex membrane curvature of phosphatidylserine (PS)-containing vesicles enhances the formation of binding sites for factor Va and lactadherin, but the relationship between convex curvature and protein localization on cells remains unknown. In this study, a membrane topology model was developed to explore this relationship further. Results show that the ridges of membranes with maximal curvature selectively bound lactadherin and factor Va, supporting the assembly of prothrombinase complexes.

This article focuses on the reasons for polyploidy, the stress responses and molecular signals involved, as well as its crucial roles in cell growth and tissue regeneration in the heart, liver, and other tissues.

Additionally, concomitant use of paclitaxel and GG genotype also showed a significant association with severe CIPN.

Summary: Extracellular vesicles (EVs) are being explored as biocompatible drug delivery vehicles for their ability to deliver bioactive cargo to recipient cells. However, effective methods for loading and unloading proteins inside EVs remain a challenge. In this study, we present the Technology Of Protein delivery through Extracellular Vesicles (TOP-EVs) as an efficient tool for intracellular protein delivery mediated by EVs. Our results demonstrate the versatility and potential of TOP-EVs for protein-based therapeutics in vitro and in vivo.

Increased Bone Marrow Uptake and Accumulation of Very-Late Antigen-4 Targeted Lipid Nanoparticles

Summary: Lipid nanoparticles (LNPs) are rapidly evolving as promising delivery systems for oligonucleotides, including siRNAs. In this study, LNPs were specifically targeted to hematopoietic progenitor cells in the bone marrow using a modified tripeptide ligand. Functionalized LNPs showed improved uptake and delivery of siRNA in patient-derived leukemia cells, as well as increased accumulation and retention in the bone marrow. These findings suggest the potential of LNPs for targeted therapy in leukemia and other hematological disorders.

Levodopa-loaded nanoparticles for the treatment of Parkinson's disease

Summary: Parkinson's disease is characterized by degeneration of dopaminergic neurons, causing dopamine deficiency and motor symptoms. Current treatment using L-DOPA has limitations and side effects. Encapsulation of L-DOPA into nanoparticles can protect it and improve delivery to the brain, reducing required dosages and side effects. Alternative delivery methods, such as intravenous administration and transdermal delivery, enhance therapeutic effects. This review discusses recent advances and future perspectives on NP-mediated L-DOPA delivery to the brain.

Nanomedicines: An approach to treat placental insufficiency and the current challenges

Summary: Preeclampsia and fetal growth restriction are pregnancy complications that can be treated with nanomedicines, which regulate drug interaction with the placenta to enhance treatment efficacy and minimize fetal exposure. Liposomes and polymeric drug delivery systems show promising results in preventing trans-placental passage of nanomedicines, while quantum dots and silicon nanoparticles have limited research in placental insufficiency syndromes. Animal studies demonstrate positive effects on maternal and fetal health, but results are influenced by various factors. Further research is needed to understand the pathophysiology of these complex diseases and ensure the safety and efficacy of nanomedicines before clinical implementation.
