Soil and spray inoculation were used to increase disease pressure and provoke genotypic differentiation. The evaluation of stable quantitative FHB resistance in barley genotypes was achieved through the quantification of fungal DNA contents and multiple Fusarium toxins. The study also identified the impact of weather conditions on fungal colonization in mature barley grain.

This study found that putrescine, a wheat defense compound, can have a significant impact on the transcriptome of F. graminearum and induce the synthesis of DON. Gene ontology and KEGG enrichment analysis revealed the involvement of various metabolic pathways. Co-expression analysis identified key transcription factor genes that may play important roles in DON synthesis. These findings provide important insights into the molecular mechanisms underlying putrescine-induced DON synthesis and facilitate the study of FHB pathogenic mechanisms.

Summary: The study explored intraspecific and interspecific diversity between Phoenix theophrasti individuals from Turkey, Crete, Greece, and P. dactylifera specimens from Turkey using ISSR analysis. Results showed that intraspecific diversity ranged from 13.45% to 26.63% for P. theophrasti and geographic and genetic distances were positively correlated. STRUCTURE analysis classified populations into four clusters, supporting the differentiation between P. theophrasti and P. dactylifera as different species.

Summary: This study investigated the antifungal effects of myrcene, a plant-based compound, on Fusarium graminearum PH-1 and Fusarium culmorum FcUK99. The minimum inhibitory concentration (MIC) and half of MIC (MIC50) of myrcene against both strains were found to be 25 μg/mL and 12.5 μg/mL, respectively. MIC50 application significantly decreased cell viabilities and increased catalase (CAT) activity. Apoptosis-like process and cellular oxidative stress were observed. Genomic template stability (GTS) percentages and methylation polymorphism values were calculated. Myrcene caused epigenetic modifications in both genomes. MIC50 dose affected the expressions of certain genes, but did not change retrotransposon movement. This study demonstrated the potential of myrcene as an effective antifungal agent with morphological, genetic, epigenetic, and cellular alterations.

Summary: The phylogenetic relationship among Fusarium graminearum Schwabe isolates was studied using mitochondrial cox3 and rnl gene variations. DNA fragments were amplified from purified isolates and sequenced. Nucleotide polymorphisms were detected by alignment and a phylogenetic relationship was constructed using PAUP 4.0a. The results showed that Korean isolates were closely related to Turkish and Iranian isolates. The methodology can be extended to other fungal species.

Summary: Heterologous expression of plant-derived SNK-Z peptide in E. coli was successfully achieved, and its tertiary structure was determined. The most efficient production of the recombinant fusion peptide (GST/SNK-Z) was achieved by induction with 0.1 mM isopropyl β-D-1-thiogalactopyranoside at 32°C. The purified GST/SNK-Z protein with a molecular weight of 30.14 kDa yielded 6.5 mg/L, and the GST tags were removed using PreScission protease. The IC50 of GST/SNK-Z for Staphylococcus aureus was calculated as 12.07 μM.
