Summary: The aim of this study was to investigate the effect of citrulline on the pyroptosis of mouse macrophage RAW264.7 and the mechanism. The results showed that citrulline promoted cell viability and inhibited LPS-induced pyroptosis of RAW264.7 cells. Furthermore, citrulline inactivated the NF-κB/p65 signaling pathway by suppressing p65 nuclear translocation induced by LPS. Therefore, the inhibition of pyroptosis induced by citrulline may be closely related to the inactivation of the NF-κB/p65 signaling pathway.

Summary: Potentilla discolor Bunge (PD) is a traditional Chinese medicine with strong anti-inflammatory effects by suppressing the expressions of iNOS, TNF-α, and MCP-1, as well as inhibiting the activation of NF-kappa B and AP-1. The ethanol extract of the whole herbs of PD (EPD) shows promising therapeutic potential for inflammatory diseases, especially intestinal inflammation, through its ability to modulate inflammatory mediators and signaling pathways.

Summary: Periodontitis is a chronic inflammatory disease caused by bacterial pathogens and the immune response. Macrophages play a crucial role in the development of periodontitis. NAT10 was found to be downregulated in macrophages during LPS-induced inflammation. Knockdown of NAT10 reduced the production of inflammatory factors, while overexpression of NAT10 had the opposite effect.

Summary: The study demonstrated that 6-MC reduced the levels of nitric oxide and prostaglandin E-2 without causing cytotoxicity, and also decreased the expression of pro-inflammatory cytokines in a concentration-dependent manner in LPS-stimulated cells. Western blot results showed that 6-MC treatment decreased the protein levels of iNOS and COX-2 induced by LPS, while mechanistic studies revealed its inhibitory effects on the MAPK and NF-kappa B pathways.
