Summary: After spinal cord injury, the activation of the Integrated Stress Response (ISR) and phosphorylation of eIF2 alpha in neurons may be a potential treatment strategy for SCI. The use of ISRIB, an ISR inhibitor, significantly reduces neuronal apoptosis, promotes morphological recovery, and enhances locomotor function following SCI.

Summary: Exposure to 2-MIB affects developmental parameters, locomotive behavior, oxidative stress, apoptosis, and transcriptome of zebrafish. However, it does not cause changes in mortality, hatchability, and malformation rate. Zebrafish larvae show increased body length in a dose-dependent manner, accompanied by alterations in GH/IGF and HPT axis genes. Additionally, 2-MIB induces increased swimming activity in zebrafish larvae.

Summary: This study found that exposure to manganese increases mortality and malformation rates in zebrafish larvae, and significantly reduces swim distance and velocity. Manganese exposure also leads to apoptosis of dopaminergic neurons and decreased expression of tyrosine hydroxylase. Transcriptome sequencing results showed upregulation of genes associated with apoptosis and DNA damage repair.

Summary: This study investigates how the RapGEF homolog PXF-1 regulates synaptic function. Loss of function mutations in pxf-1 decrease cholinergic activity at the neuromuscular junction, resulting in reduced levels of synaptic vesicles and filamentous actin. PXF-1 promotes the formation of actin filaments to support the development of motor neuron synapses.

Summary: In this study, it was found that TAK1 is involved in neuronal survival in the cerebral cortex and hippocampus of rats with chronic hypertension. TAK1 can suppress apoptosis and necroptosis, improving cognitive function. This result was verified in vitro. This study provides new insights into the cognitive impairment caused by chronic hypertension.

Summary: The pathogenesis of Parkinson's disease (PD) is complex and requires further exploration. Leucine-rich repeat kinase 2 (LRRK2) is associated with both familial and sporadic PD. Abnormal iron accumulation is found in the substantia nigra of PD patients, but its exact effects are not clear. This study shows that iron dextran exacerbates neurological deficits and loss of dopaminergic neurons in rats with 6-OHDA lesions, and iron promotes the activation of LRRK2, which in turn accelerates ferrous iron uptake.

Summary: This study evaluated the potential toxicity of four commonly used strobilurin fungicides on larval zebrafish, finding that pyraclostrobin and trifloxystrobin induced mitochondrial dysfunction and hypoactive responses in zebrafish. However, azoxystrobin and kresoxim-methyl had no significant effects on mitochondrial function or locomotion in zebrafish. The findings suggest a potential for adverse effects in aquatic organisms due to widespread detection of strobilurins in aquatic environments.

Summary: In this study, the transcriptomic signatures in POCD mice were analyzed to identify the expression profiles of circRNA, miRNA, and mRNA. A ceRNA regulatory network was established, revealing the potential role of circRNA, miRNA, and mRNA interactions in POCD. These findings suggest that circ_0001634/miR-490-5p/Rbm47, circ_0001634/miR-490-5p/Sostdc1, circ_0001634/miR-7001-5p/Sostdc1, circ_0001345/miR-7001-5p/Sostdc1, and circ_0001493/miR-7001-5p/Sostdc1 may serve as novel diagnostic biomarkers and therapeutic targets for POCD.

Summary: Activation of NCOA4-mediated ferritinophagy exacerbates early-stage CIRI and plays a crucial role in the progression of oxidative stress, autophagy, and apoptosis. The cGAS-STING pathway is implicated in regulating NCOA4-mediated ferritinophagy, and its inhibition ameliorates CIRI, attenuated by NCOA4 overexpression.

Summary: This study found that PINK1-mediated mitophagy and caspase-3/GSDME pathway play important roles in the pathogenesis of postoperative cognitive dysfunction (POCD). Interventional studies using PINK1 silencing and caspase-3 inhibitor showed that PINK1-regulated mitophagy regulates anesthesia and surgery-induced cognitive impairment by affecting the caspase-3/GSDME pathway, providing a promising therapeutic target for POCD.

Summary: This study investigates the role of GRSF1 in cerebral ischemia-reperfusion injury (CIRI) and its specific mechanisms. The researchers found that GRSF1 expression was low in CIR mice, while senescence-related markers were increased. Overexpression of GRSF1 improved infarct volume and neurological function, and suppressed apoptosis. The results suggest that cellular senescence contributes to CIRI, and GRSF1 can protect against reperfusion injury by regulating GPX4.

Summary: PTEN-induced kinase 1 (PINK1)-mediated mitophagy and caspase-1/gasdermin D canonical pyroptosis pathways are involved in the development of postoperative cognitive dysfunction (POCD). Gasdermin E (GSDME), which can be activated by caspase-3, is highly expressed in the brain and neurons. This study investigates the role of PINK1-dependent mitophagy in postoperative cognitive impairment through caspase-3/GSDME.
