Summary: This study investigates the prognostic value of transcriptomes associated with tumor FDG uptake in hepatocellular carcinoma (HCC). Through analyzing the correlations between metabolic genes and FDG uptake, multiple genes related to glycolysis and fatty acid metabolism were identified. A metabolic gene expression balance score was established and showed good prognostic value in HCC. These findings highlight the potential use of FDG PET/CT imaging biomarkers in selecting appropriate patients for metabolism-targeted therapy in HCC.

Summary: This study explored the expression and function of 40S ribosomal protein S24 (RPS24) in hepatocellular carcinoma (HCC). Results showed that RPS24 was elevated in HCC samples and related to poor prognosis. Furthermore, RPS24 overexpression or promoter methylation correlated with unfavorable prognosis in HCC patients. High expression of RPS24 was associated with DNA replication, cell cycle, immune infiltration, and immunotherapy response. Experimental validation demonstrated that silencing of RPS24 suppressed HCC cell growth and tumor proliferation. Therefore, RPS24 may serve as a potential adverse biomarker for HCC prognosis, acting through cell proliferation facilitation and the formation of an immunosuppressive microenvironment, and targeting RPS24 could be a promising therapeutic option for HCC management.

Summary: This study investigated the expression of GPBAR1 in HCC tissues and tumor-adjacent tissues and found that GPBAR1 was highly expressed in HCC tissues. GPBAR1 expression was significantly associated with poor prognosis in HCC and served as an independent prognostic biomarker. Additionally, GPBAR1 expression was significantly correlated with asynchronous bone metastasis but not liver or lung metastases. These findings provide new insights into HCC metastasis and improve prognostic assessment of HCC.

Summary: The study established a prognostic biomarker based on selenium metabolism regulators for hepatocellular carcinoma (HCC) and validated the role of INMT in HCC. A selenium metabolism model containing INMT and SEPSECS was constructed, which was shown to be an independent predictor of prognosis. The low-risk group had significantly longer survival time compared to the high-risk group, and these two groups exhibited different immune environments. INMT was significantly downregulated in HCC tissues in different datasets, and knockdown of INMT promoted HCC cell proliferation.

Summary: Immune escape and metabolic reprogramming are important characteristics of tumor biology in hepatocellular carcinoma (HCC). This study establishes a prognostic signature based on immune- and metabolism-related genes (IMRGs) to predict prognosis, evaluate immunotherapy efficacy, and personalize immunotherapy for HCC patients. The signature shows superior performance compared to previously published HCC signatures and is validated in internal and external cohorts. A nomogram combining the IMRGs signature with clinical parameters is also developed to predict prognostic risk.

Additionally, the activation of STAT3 signaling is observed in SHC4 overexpressed HCC cells and tissues. Moreover, intervention of STAT3 confirms its importance in the oncogenic role of SHC4 in HCC.
