Summary: An efficient two-step procedure for synthesizing pyrimidine nucleosides is described. Firstly, a series of glycosyl 5-(aminomethylene)-1,3-dioxane-4,6-dione derivatives were prepared, followed by transacylation reaction to obtain uridine-5-carboxylic acid derivatives and related nucleosides.

Summary: Peptidomimetics with a substituted imidazo[1,2-a ]pyridine fragment were synthesized using a combination of Groebke-Black-burn-Bienayme and Ugi reactions. The resulting products contained substituted imidazo[1,2-a]pyridine and peptidomimetic moieties as pharmacophores, with four diversity points introduced from readily available starting materials. A focused compound library consisting of 20 Ugi products was prepared and screened for antibacterial activity.

Summary: Meldrum's acid is a molecule with exceptional chemical properties, characterized by its low pK(a) value in water and reactivity in electrophilic and nucleophilic substitution reactions. At elevated temperatures, it undergoes distinctive decomposition pathways that can be used in cycloaddition and acylation reactions. Recent advances in utilizing Meldrum's acid derivatives in synthesis aim to further stimulate research towards innovative applications of this molecule.

Summary: We have developed an efficient modular asymmetric synthesis of azahelicenes from readily available polycyclic aromatic amines, aldehydes, and (di)enamides, employing a central-to-helical chirality conversion strategy. A series of aza[5]- and aza[4]helicenes bearing various substituents were easily obtained with good yields and high enantioselectivities. The diverse derivatizations and the preliminary application of the azahelicene derivative as a chiral organocatalyst demonstrated the potential of this method, and the photophysical and chiroptical properties of these azahelicenes were well studied, showing potential applications in novel organic optoelectronic materials.

Summary: We have discovered biosynthetic pathways that can produce and transform conjugated octaenes and nonaenes into complex natural products. The biosynthesis of (-)-PF1018 involves an enzyme called PfB, which controls the regio-, stereo-, and periselectivity of multiple reactions starting from a conjugated octaene. By using PfB as a starting point, we found a homologous enzyme, BruB, that facilitates diene isomerization, tandem 8π-6π-electrocyclization, and a 1,2-divinylcyclobutane Cope rearrangement to generate a novel compound not previously found in nature.

Summary: The effective preparation of two new pyrimidine- and pyridine-based organic crystalline salts with substituted acidic moieties using methanol as a solvent has been reported. These molecular salts have ionic interactions responsible for their structural stabilization in solid-state assemblies. The crystal structures of the salts were determined and their noncovalent interactions were investigated.
