Summary: We discovered a novel mechanism of ErbB2-driven 3D breast tumor growth mediated by ErbB2-dependent BLNK downregulation. In addition, we found that upregulating BLNK can inhibit tumor growth, and increased mRNA levels of BLNK are associated with increased relapse-free survival.

Summary: This study aimed to quantify the amino acid levels in cultured fibroblasts and compare different sample-based normalization approaches. The results showed that normalizing amino acid values by phenylalanine can achieve the lowest variability and identify different fibroblast groups. Therefore, phenylalanine may be a suitable biomarker to estimate cellular content in cultured fibroblasts.

Summary: In this study, the syntheses, characterisation, properties comparison, and applications of molybdate and polyoxomolybdate ionic liquids as bifunctional catalysts for CO2 insertion into epoxides were described. The synthetic methods relied on anion exchange and acid-base reactions, including a novel route for the synthesis of molybdate ionic liquids using a halide-free organic precursor. The investigations showed that tetrabutylammonium molybdate could serve as an efficient catalyst for the production of hexane carbonate from 1,2-epoxyhexane under solventless and halide-free conditions.

Summary: This study identified an IL-22-dependent gene signature in a human intestinal cell line, which could serve as a pharmacodynamic biomarker for IL-22 therapy. HT-29 cells demonstrated dose-dependent increases in STAT3 phosphorylation and multiple gene expression changes in response to UTTR1147A.

Summary: This paper proposes a quantitative identification method based on bounded component analysis (BCA) for both independent and dependent vibration sources in mechanical systems. The method separates source signals, extracts source contribution, and calculates the projection of source contribution in mixed signals to evaluate the influence of vibration sources. Simulation analysis and experimental verification show high accuracy in the quantitative identification of both independent and dependent sources.

Summary: Immunotherapy using antigen-specific antibodies or immune checkpoint inhibitors has greatly impacted breast cancer treatment. A new image-based assay has been developed to quantify antibody-dependent cellular cytotoxicity (ADCC) and identify novel ADCC modulator compounds. This assay can be used to screen compound libraries and identify compounds that enhance ADCC, potentially serving as adjuvant therapeutic agents for breast cancer immunotherapy.

Summary: Through knockout of key proteins MSH2, MSH3, and MSH6 in homozygous state, it is revealed that MutSa and MutSss determine the efficiency of prime editing in a size-dependent manner in human pluripotent stem cells (hPSCs). MSH2 perturbation significantly escalated prime editing efficiency from base mispair to 10 bases, while impaired MutSa and MutSss by MSH6 and MSH3 knockout respectively improved editing efficiency in different size ranges. This study highlights the importance of mismatch repair (MMR) in determining prime editing efficiency in hPSCs and demonstrates the distinct roles of MutSa and MutSss in its outcome.

Summary: The decreased functionality and expression of HpHbR in Trypanosoma brucei gambiense is a key evolutionary modification that allows this parasite to infect humans. This study reveals that hemoglobin uptake in African trypanosomes is almost exclusively mediated by HpHbR and crucial for their development. Understanding this molecular mechanism is important for our knowledge of how the parasite infects humans and for the development of potential treatments.

Summary: This study isolated Caenibius tardaugens for its ability to degrade estrogenic endocrine disruptors and identified the regulatory role of EdcR in the upper degradation pathway of 17 beta-estradiol. The findings of genetic studies also suggest the horizontal transfer of this regulatory system to other bacteria.
