Summary: A novel tT beta RII variant, Z-tT beta RII, was designed by fusing the PDGF beta receptor-specific affibody Z(PDGF beta R) to the N-terminus of tT beta RII. Z-tT beta RII exhibited superior specific fibrotic liver-targeting potential and stronger anti-fibrotic effects in vitro and in vivo. It also showed no significant sign of potential side effects in other vital organs in liver fibrotic mice, making it a potential candidate for targeted therapy for liver fibrosis.

Summary: Human epidermal growth factor receptor-2 (HER2) is a well-known cancer target with various therapies available. However, the response and effectiveness of HER2-targeted agents differ among different tumor types, including gastric adenocarcinomas (GACs). GACs exhibit heterogeneity in HER2 expression, both within and between tumors, which poses challenges in assessing the efficacy of HER2-targeted agents. The recent approval of trastuzumab deruxtecan, an antibody drug conjugate, for refractory HER2-positive advanced GAC patients shows promise. Ongoing research aims to understand resistance patterns and intra-tumor heterogeneity to improve treatment outcomes. This review discusses the current status of HER2-targeted therapy in GACs and explores new therapies under investigation.

Summary: Epithelial-to-mesenchymal transition (EMT) is a biological process that promotes cancer cell dissemination and drug resistance. This study revealed that overexpression of MIR31HG in pancreatic ductal adenocarcinoma (PDAC) patients is associated with poorer disease-free survival, as well as upregulation of genes related to TGF beta signaling and EMT. In vitro experiments further demonstrated that TGF beta induces MIR31HG expression and knockdown of MIR31HG reverses TGF beta-induced EMT phenotypes and cancer cell migration.
