Epigenetic modifications and NF-κB pathway activity in Cu,Zn-SOD-deficient mice

The aim of this study was to examine the possible impact of Cu,Zn-SOD deficiency on the level of epigenetic modifications in different mouse tissues, and the relationship between these modifications and the NF-kappa B transcription factor activity. Cu,Zn-SOD deficiency did not influence the level of 5mdC or 5hmdC in the analyzed tissues. Statistically significant organ-/tissue-specific differences between the levels of 5mdC and 5hmdC were demonstrated within each genotype. Also correlations between analyzed parameters pointed to wide tissue/genotype variety; we observed a positive correlation between 5mdC and NF-(DB)-B-0 proteins, p50 and RelA, in the liver of wild mice, as well as an inverse correlation between 5mdC and p65 in the brain of Cu,Zn-SOD-deficient animals. Moreover, a positive correlation was revealed between 5mdC and 5hmdC in the liver and brain of knockout mice. As the highest levels of both 5mdC and 5hmdC were observed in the brains of analyzed animals regardless of their genotype, and lower, comparable to each other, levels of these modifications were shown in the kidney and liver, active demethylation process seems to be tissue-/organ-specific and does not necessarily rely solely on the redox/oxidation state of cells. According to the most likely scenario, various tissues may differ in terms of their metabolic rates, which has potential influence on cofactors, and consequently on the activity of TET enzymes or activation of TET-independent mechanisms.

An IDH-independent mechanism of DNA hypermethylation upon VHL inactivation in cancer

Summary: DNA methylation plays a significant role in cancer progression and can be affected by various environmental conditions, such as hypoxia. In renal cancer cells, inactivation of the VHL gene under normoxic conditions induces global genome hypermethylation, which can be reversed by exogenous expression of wild-type VHL. The hypermethylation in VHL mutants is caused by transcriptional changes in MDH and L2HGDH genes leading to accumulation of 2-hydroxyglutarate, inhibiting DNA demethylation by TET enzymes.

Expression of Iron Metabolism Proteins in Patients with Chronic Heart Failure

Summary: This study investigated the changes in iron-related proteins in the myocardium of heart failure patients. The results showed reduced expression of several proteins responsible for iron transport and storage, along with increased expression of oxidative stress markers. The reduction of iron-gathering proteins and elevated oxidative stress in failing hearts is detrimental to myocardial cells.

Silver Nanoparticles Inhibit Metastasis of 4T1 Tumor in Mice after Intragastric but Not Intravenous Administration

Summary: This study analyzed the impact of different silver nanoparticles on the development and metastatic potential of breast cancer using a mouse model. The results showed that citrate-coated silver nanoparticles had a significant effect on reducing tumor volume and lung metastases in mice treated intragastrically. However, no effect on lung metastases was observed after intravenous administration of silver nanoparticles. The findings suggest that the anticancer activity of silver nanoparticles may depend on the activation of the immune system.

Summary: Heart failure (HF) is a common disease and iron deficiency (ID) plays a significant role in worsening the condition of HF patients. Iron supplementation is recommended but may have adverse effects. There are still many knowledge gaps regarding the interaction between iron deficiency and HF. Summarizing the latest translational and clinical research on iron deficiency in HF is important for raising awareness among clinicians.

Global DNA 5-hydroxymethylcytosine level and its chromosomal distribution in four rye species

Silver Nanoparticles Induced Changes in DNA Methylation and Histone H3 Methylation in a Mouse Model of Breast Cancer

Summary: The epigenetic effects induced by citrate- and PEG-coated silver nanoparticles (AgNPs) in a 4T1 breast cancer mouse model were analyzed. It was found that both types of AgNPs significantly decreased the level of 5-methylcytosine (5-mC) in tumor tissue, with the effect being more pronounced for citrate-coated AgNPs. PEG-coated AgNPs also caused a significant decrease in DNA methylation after intravenous administration, and both types of AgNPs led to a decrease in histone H3 methylation.

Exposure to Chlorpyrifos Alters Proliferation, Differentiation and Fatty Acid Uptake in 3T3-L1 Cells

Summary: Chlorpyrifos, a representative of organophosphorus pesticides, has effects on the viability, proliferation, differentiation, and fatty acid uptake in 3T3-L1 cells. Exposure to chlorpyrifos decreases cell viability but increases it at low concentrations. It inhibits proliferation and differentiation of preadipocytes, reduces lipid accumulation, and down-regulates key transcription factors. In fully differentiated adipocytes, chlorpyrifos increases basal free fatty acid uptake. These findings suggest that chlorpyrifos may play a role in the development of obesity.

Adverse Effects of Non-Metallic Nanoparticles in the Central Nervous System

Summary: The interest in nanoparticles and their effects on living organisms has been increasing in recent decades. Non-metallic nanoparticles appear to be less toxic than metallic ones, leading to their growing use in medicine and industry. This review summarizes current knowledge of the in vitro and in vivo neurotoxicity of non-metallic nanoparticles, as well as the mechanisms associated with their positive or negative effects on the central nervous system (CNS).

Polystyrene nanoparticles: the mechanism of their genotoxicity in human peripheral blood mononuclear cells

Summary: This study investigated the genotoxic potential of non-functionalized polystyrene nanoparticles (PS-NPs) of different diameters on human peripheral blood mononuclear cells (PBMCs). The results showed that PS-NPs decreased the metabolic activity of PBMCs, increased DNA strand break formation, oxidized purines and pyrimidines, and increased 8-oxodG levels. The extent of genotoxic changes in PBMCs depended on the size of the particles and their zeta-potential value.
