Summary: Replacing silver metallization with earth-abundant materials is an important step for sustainable growth of photovoltaics. In this study, fire-through processes using aluminum as the metallization material for solar cells were investigated. It was found that increasing the nitrogen content of the silicon nitride can reduce aluminum penetration depth. The mechanisms behind metal-induced passivation degradation were also investigated and possible mitigation strategies were identified using an adapted SiOx/SiCx stack for tunnel oxide passivating contacts.

Summary: This study demonstrates that mitochondrial DNA stress upregulates the immunoproteasome and MHC class I antigen presentation pathway via cGAS/STING/type I interferon signaling, leading to cell autonomous activation of CD8(+) T cells. This response is observed in both mice and humans and is associated with CD8(+) T-cell activation in lung epithelial cells of patients with idiopathic pulmonary fibrosis. Genetic depletion of the immunoproteasome and specific immunoproteasome inhibitors counteract DNA stress-induced cytotoxic CD8(+) T-cell activation. These findings highlight the role of cytoplasmic DNA sensing through the cGAS/STING pathway in activating the immunoproteasome and CD8(+) T cells, providing a potential new therapeutic target for pulmonary fibrosis.

Summary: The acquisition of mesenchymal traits is a key factor in breast cancer progression. However, the relevance of epithelial-to-mesenchymal transition (EMT) is controversial and depends on the specific context. In this study, researchers isolated epithelial and mesenchymal populations from metastatic breast cancer biopsies and assessed their functional potential in vivo. The results show that decreasing levels of the epithelial cell adhesion molecule (EPCAM) are associated with a decline in disease propagation. Mechanistically, persistent EPCAM expression identifies epithelial clones that resist EMT induction and propagate competitively, while loss of EPCAM defines clones arrested in a mesenchymal state with suppressed tumorigenicity and metastatic potential. These findings highlight the importance of EMT in breast cancer metastasis and suggest potential targets for therapeutic intervention.

Summary: Interstitial lung disease (ILD) is a diverse group of lung disorders characterized by persistent architectural distortion of the lung parenchyma. In this study, a 3D organoid co-culture model was developed to investigate the mesenchymal-epithelial crosstalk in ILD. The results showed that ILD fibroblasts increased metabolic activity and altered stem cell function in alveolar organoids.

The impact of the immune system on lung injury and regeneration in COPD

Theodore S. Kapellos, Thomas M. Conlon, Ali oender Yildirim, Mareike Lehmann

Summary: This review provides recent evidence on the contribution of immune cells to lung injury and regeneration. The article focuses on four main aspects: the mechanisms of alveolar degradation caused by myeloid cells, the formation of tertiary lymphoid structures and production of autoreactive antibodies, the consequences of inefficient apoptotic cell removal, and the effects of innate and adaptive immune cell signaling on alveolar epithelial proliferation and differentiation. The article also discusses how recent technological advances in omics technologies and human ex vivo lung models can elucidate the interaction between immune cells and epithelial cells and expedite precision pro-regenerative approaches to treat COPD.

Summary: This study evaluated the prevalence of tooth agenesis in German orthodontic patients with non-syndromic cleft lip and/or palate and Robin sequence compared to a control group without craniofacial disorder. The results showed a statistically significant higher prevalence of tooth agenesis in patients with non-syndromic craniofacial disorder.
