The association between fecal microbiota and different types of colorectal polyp as precursors of colorectal cancer

Background: Colon cancer (CRC) is a heterogeneous disorder, arising from precursors-adenoma and serrated polyp. Previous studies have demonstrated the relationship between the human gut microbiota and CRC; however, its correlation to the different early precursors of CRC is not properly understood. Here, we studied the relationship between targeted gut bacteria and different colorectal polyp types, location, size and grade of dysplasia. Material and methods: In the present case-control descriptive study, selected fecal bacteria were assessed in 118 patients, referred for standard screening colonoscopy, including 31 normal controls, 21 hyperplastic polyp (HP), 16 sessile serrated polyp (SSA), 29 tubular adenoma (TA) and 21 villous/tubuvillous polyp (VP/TVP) cases, between 2015 and 2017, by absolute quantitative real time PCR technique (q PCR) in different ethnicity of Iranian population. The panel of bacteria was including Streptococcus bovis/gallolyticus, Enterococcus faecalis, Enterotoxigenic Bacteroides fragilis (ETBF), Fusobacterium nucleatum, Porphyromonas spp., Lactobacillus spp., Roseburia spp., and Bifidobacterium spp. Results: Higher numbers of F. nucleatum, E. feacalis, S. bovis, ETBF and Porphyromonas spp. were detected in AP cases, consisting TA and especially VP/TVP, in contrast to samples from the normal, HP and SSA groups (P < 0.001). On the contrary, lower number of Lactobacillus spp., Roseburia spp. and Bifidobacterium spp. were detected in AP, compared to the normal, HP and SSA. Surprisingly, a significant correlation was found among selected gut bacterial quantity, the size, location and grade of dysplasia of polyp cases. Discussion: These findings suggest that gut bacteria might contribute in early stages of colorectal carcinogenesis through the development of AP, but not SSA. In fact, AP and SSA are also different in terms of molecular pathways and tendencies to present in specific colorectal location. Overall, these findings may lead to development of CRC prevention therapies, targeting early protagonist bacteria of colorectal carcinogenesis from AP.

Summary: This study conducted monitoring of SARS-CoV-2 in sewage from seven wastewater treatment plants in Tehran, Iran and investigated the correlation between viral loads in sewage and infected populations. The results showed a positive correlation between viral load in sewage and the outbreak of COVID-19, with the presence of SARS-CoV-2 RNA in sewage predicting population infections 1-2 days in advance.

Summary: This study investigated the association between CTGF and MMP-1 mRNA expressions with clinicopathological status and survival rate in colorectal cancer patients. The results showed higher expression levels of MMP-1 and CTGF in tumors compared to adjacent normal tissues, and their overexpression was associated with lymph node metastasis, distant metastasis, advanced stage, and poor survival.

Summary: This study examined the temporal patterns of colorectal cancer burden and its risk factors globally, regionally, and nationally over the past three decades. The findings showed a continuous increase in incidence and mortality of colorectal cancer globally, with rising rates in younger adults and a significant burden in Asian and African countries. Diet, smoking, and alcohol use were identified as major contributors to colorectal cancer burden.

Fanconi Anemia Pathway in Colorectal Cancer: A Novel Opportunity for Diagnosis, Prognosis and Therapy

Summary: Colorectal cancer (CRC) is the third most commonly diagnosed malignancy globally. This article explores the role of Fanconi anemia (FA) genes in CRC susceptibility, as mutations in these genes increase the risk of tumor development. The FA pathway is involved in DNA repair and genome stability, and has been associated with CRC progression and drug resistance. FANC genes have the potential to serve as promising prognostic and predictive biomarkers in CRC management and treatment, as well as targets for precision cancer medicine.

Summary: Colorectal cancer, the second leading cause of cancer death worldwide, is closely associated with epigenetic changes such as DNA methylation. Studies have shown that DNA methylation markers have high accuracy and reproducibility in noninvasive biosamples, making them promising for early diagnosis and personalized treatment of CRC. Further research and development of methylation biomarkers and detection methods are crucial for CRC diagnosis.

Summary: On 25 November 2021, the World Health Organization identified Omicron as a newly emerged and the fifth Variant of Concern (VoC) of SARS-CoV-2. Omicron has the highest number of mutations compared to other VoCs and is the only variant with changes in the receptor-binding domain (RBD). Despite being detectable with previous PCR tests, Omicron has a higher transmission rate and poses challenges to immune response and vaccine effectiveness.

Identification of LncPVT1 and CircPVT1 as prognostic biomarkers in human colorectal polyps

Integrated bioinformatics and wet-lab analysis revealed cell adhesion prominent genes CDC42, TAGLN and GSN as prognostic biomarkers in colonic-polyp lesions

Summary: This study investigated the expression pattern of CDC42, TAGLN, and GSN genes in high-risk and low-risk polyps, as well as colorectal cancer patients and their adjacent normal tissues. The results showed that these genes were significantly elevated in high-risk adenomas and associated with clinicopathological characteristics. The expression of CDC42 and TAGLN genes was significantly related to survival rates, suggesting their potential as diagnostic or prognostic markers for colorectal cancer.
