Recognition of enteroinvasive Escherichia coli and Shigella flexneri by dendritic cells: distinct dendritic cell activation states

The innate and adaptive immune responses of dendritic cells (DCs) to enteroinvasive Escherichia coli (EIEC) infection were compared with DC responses to Shigella flexneri infection. EIEC triggered DCs to produce interleukin (IL)-10, IL-12 and tumour necrosis factor (TNF)-alpha, whereas S. flexneri induced only the production of TNF-alpha. Unlike S. flexneri, EIEC strongly increased the expression of toll like receptor (TLR)-4 and TLR-5 in DCs and diminished the expression of co-stimulatory molecules that may cooperate to inhibit CD4(+) T-lymphocyte proliferation. The inflammation elicited by EIEC seems to be related to innate immunity both because of the aforementioned results and because only EIEC were able to stimulate DC transmigration across polarised Caco-2 cell monolayers, a mechanism likely to be associated with the secretion of CC chemokine ligands (CCL) 20 and TNF-alpha. Understanding intestinal DC biology is critical to unravelling the infection strategies of EIEC and may aid in the design of treatments for infectious diseases.

Summary: Sporotrichosis is a subcutaneous mycosis distributed worldwide, mainly caused by traumatic inoculation of propagules. Research has shown that extracellular vesicles from Sporothrix brasiliensis play a protective role during infections by increasing fungicidal activity and elevating certain interleukin levels in mice. This indicates a potential therapeutic approach in utilizing EVs for protective effects during Sporothrix infections.

Summary: This study compared the performance of three different mRNA vaccine modalities in targeting tumors associated with HPV-16 infection in mice. The results showed that all three vaccines effectively activated E7-specific CD8+ T cells, generated memory T cell responses, prevented tumor relapses, and eradicated subcutaneous tumors at different growth stages. In addition, the mRNA-LNP vaccines showed potent tumor protection in two different mouse tumor models. Compared to DNA and recombinant protein vaccines, the mRNA vaccines demonstrated superior immunogenicity and therapeutic efficacy. These findings support further evaluation of these mRNA vaccines in clinical trials.

Targeting the P10 Peptide in Maturing Dendritic Cells via the DEC205 Receptor In Vivo: A New Therapeutic Strategy against Paracoccidioidomycosis

Summary: Paracoccidioidomycosis (PCM) is a systemic mycosis caused by Paracoccidioides brasiliensis, a common endemic fungus in Latin American countries. Vaccines targeting strong T cell-mediated immune responses are being developed. The P10 peptide derived from the fungus has shown promise in inducing immune responses when targeted to dendritic cells (DC) with a monoclonal antibody to the DEC205 receptor. In experiments with mice, treatment with the aDEC/P10 chimeric antibody resulted in increased IFN-γ and IL-4 levels, reduced fungal burdens, and normal lung tissue architecture. This strategy shows potential for vaccination and therapy against PCM.

Summary: Traditional lab classes in microbiology are common in educational institutions. In order to provide a more authentic learning experience, the Real-Lab-Day was developed as a multimodal learning experience for undergraduate students to develop competencies, critical analysis, and teamwork skills. Students were mentored by graduate students in research laboratories and conducted scientific assays to address questions in microbiology. The Real-Lab-Day improved students' learning and interest in microbiology research, and was highly approved as a teaching tool.
