Platelet-rich plasma (PRP) is a plasma fraction in which several growth factors are concentrated at high levels. In recent years, the biological effects on various cells of the active soluble releasate that is isolated following platelet activation of PRP [PRP-releasate (PRPr)] have been reported. The purpose of this study was to determine the angiogenic effects of human PRPr in vitro and in vivo. PRPr was prepared from human whole blood using the double spin method and was activated with CaCl2 and autologous thrombin. PRPr stimulated proliferation, migration and tube formation of human umbilical vein endothelial cells (HUVECs) and in vivo angiogenesis-inducing ability in nude mice. PRPr led to the phosphorylation of Erk1/2 and Akt in HUVECs, and the induction of proliferation and migration by PRPr was suppressed by PRPr inhibitors PD98059 and LY294002. PRPr induces angiogenesis in vitro and in vivo, and the present findings suggest that the mechanism for this is activation of the ERK and phosphatidylinositol-3-kinase-Akt pathways. Our results demonstrate that PRPr is a promising autologous source for therapeutic angiogenesis in treating cardiovascular disease.

Application of geometric morphometrics for facial congenital anomaly studies

Motoki Katsube, Shigehito Yamada, Natsuko Utsunomiya, Naoki Morimoto

Summary: The face is a small complex three-dimensional structure composed of various bones and essential organs. Geometric morphometrics (GM) uses shape configurations to retain geometric information and help visualize results, making it advantageous in morphometric studies of congenital facial anomalies.

Treatment of giant congenital melanocytic nevi with cultured epithelial autografts: Clinical and histopathological analysis

A. Shoji-Pietraszkiewicz, M. Sakamoto, M. Katsube, S. Ogino, I Tsuge, H. Yamanaka, J. Arata, N. Morimoto

Summary: In this study, cultured epithelial autografts (CEA) were applied after curettage or abrasion of giant congenital melanocytic nevi (GCMN). The results showed successful growth of CEA on wounds and improvement in lesion color. Histopathological examination revealed regenerated dermis without nevus cells.

Summary: CEA appears to accelerate epithelization after curettage or ablation of CMN, and can significantly improve the color of CMN when applied initially. The color of the target nevus improved significantly in both the Curettage group and non-Curettage group with initial treatment, with no significant differences in epithelization period or incidence of hypertrophic scars among the groups.

Development of gelatin hydrogel nonwoven fabrics (Genocel®) as a novel skin substitute in murine skin defects

Yuanjiaozi Li, Eiichi Sawaragi, Michiharu Sakamoto, Takashi Nakano, Hiroki Yamanaka, Itaru Tsuge, Kumiko Matsuno, Yasuhiko Tabata, Naoki Morimoto

Summary: The study concluded that Genocel (R) can serve as an equivalent skin substitute for wound healing, demonstrating comparable performance to conventional substitutes. Additionally, Genocel (R) exhibited the ability to accelerate capillary formation, making it a more suitable treatment option for chronic skin ulcers such as diabetic ulcers.

Dried human-cultured epidermis accelerates wound healing in a porcine partial-thickness skin defect model

Summary: Autologous cultured epidermis (CE) is an effective approach for treating extensive burns, but its production time is too long. Allogeneic CE can be prepared in advance and used as a wound dressing to promote wound healing. Dried CE, prepared by removing all water and viable cells, accelerates wound healing and may be a new therapeutic strategy. This study evaluated the safety and efficacy of dried CE in wound healing using a miniature swine model.

The volume-outcome relationship in free-flap reconstruction: A nationwide study based on the Clinical Database

Shinsuke Akita, Hiraku Kumamaru, Hisashi Motomura, Nobuyuki Mitsukawa, Naoki Morimoto, Minoru Sakuraba

Summary: This study analyzed data from 19,482 free-flap surgeries performed at 407 facilities in Japan and found that free-flap surgeries performed at institutions with a low average annual number of procedures had a higher risk of total necrosis. It is recommended to either aggregate cases into high-volume centers or improve management at low-volume centers.

Summary: This study successfully developed a self-assembled, cell-based dermal substitute derived from human dermal fibroblasts through long-term three-dimensional culture. The scaffold-free cultured dermis exhibited superior properties, such as sufficient tensile strength, high biocompatibility, and efficacy in promoting wound healing when compared to the collagen-based artificial dermis.
