Summary: Hypoxia and increased levels of inflammatory cytokines in the joints are characteristics of rheumatoid arthritis (RA). This study investigated the effects of hypoxia and tumor necrosis factor-alpha (TNF-alpha) on the expression of interleukin (IL)-6 and IL-8 in human fibroblast-like synoviocytes (FLSs) isolated from RA patients. Hypoxia or TNF-alpha treatment alone increased the expression and promoter activity of IL-6, IL-8, and hypoxia-inducible factor-1 alpha (HIF-1 alpha). Treatment of hypoxic FLSs with TNF-alpha further elevated the expression of these cytokines and enhanced promoter activity of HIF-1 alpha. Additionally, the combination of hypoxia and TNF-alpha increased IL-6 and IL-8 expression via the TAK1/NF-kappa B/HIF-1 alpha signaling pathway.

Summary: By comparing post-mortem lung samples from patients who died of severe COVID-19 and H1N1pdm09 infection, it was found that the H1N1 group showed an increase in tissue expression of IL-8/IL-17A and a higher number of neutrophils. There was no significant difference in the distribution of genotype frequencies in the IL17A gene between the two groups. However, in SNP rs3819025 (G/A), the G allele may be considered a risk allele in patients who died from COVID-19.

Summary: Bladder cancer is the 10th most common form of cancer globally, and chronic inflammation may play a role in its development and progression. This study aimed to investigate the association between selected single nucleotide polymorphisms (SNPs) in IL-6 and IL-8 genes and bladder cancer development, as well as the impact of bladder cancer on the expression and methylation of IL-6 and IL-8 promoters in PBMCs. The study found no association between the studied SNPs and bladder cancer occurrence. However, bladder cancer patients had decreased IL-6 and IL-8 mRNA expression levels and higher methylation status of the IL-6 promoter compared to controls.

Summary: Mammary gland tumors are a common malignant disease that poses a serious threat to women and female dogs. The lack of an effective tumor marker hinders the early diagnosis and prognosis of these tumors in veterinary clinical medicine. This study investigated the expression of IL-6, IL-8, and IL-10 in canine mammary gland tumors to determine their association with tumor progression. The results suggest that IL-6 and IL-8 could potentially serve as diagnostic or biomarkers for canine mammary gland tumors.
