Contents This study was undertaken to investigate differences in protein expression between high- and low-motility sperm of swamp buffalo. The research used two-dimensional gel electrophoresis (2DE) coupled to matrix-assisted laser desorption/ionization time-of-flight tandem mass spectrometry (MALDI-TOF/TOF-MS) to analyse the different proteins. The results showed 18 different expression protein spots between high- and low-motility buffalo sperm; eight of these proteins were up-regulated in low-motility sperm, five were down-regulated, one deleted and four proteins specifically expressed. Finally, four proteins were successfully identified by MS as belonging to three unique proteins; they are outer dense fibre of sperm tails protein 2 (ODF2), ATP synthase subunit alpha (ATP5A1) and succinyl-CoA synthetase subunit beta (SUCLG2). In summary, these results help to develop an understanding of the molecular mechanisms associated with low-motility sperm and provide clues for finding molecular markers associated with sperm motility.

Summary: This study conducted an integrative analysis of the proteome and phosphoproteome of Sertoli cells in buffalo testes to explore their role in spermatogenesis. Multiple proteins were identified, with some being involved in gene expression, cellular localization, and transport processes. The results offer insights into male reproduction and provide a foundation for future research.

Comparative transcriptome analysis reveals potential testosterone function-related regulatory genes/pathways of Leydig cells in immature and mature buffalo (Bubalus bubalis) testes

Liangfeng Huang, Kai Xiao, Junjun Zhang, Pengfei Zhang, Wengtan He, Yuyan Tang, Weihan Yang, Xingchen Huang, Runfeng Liu, Xianwei Liang, Xingting Liu, Qiang Fu, Yangqing Lu, Ming Zhang

Summary: This study analyzed the transcriptome profiles of immature Leydig cells (ILC) and adult Leydig cells (ALC) in buffalo, identifying key genes and pathways associated with the regulation of testosterone secretion function. The findings suggest that cAMP signaling plays a crucial role in maintaining testosterone function during postnatal development of LCs, with ALCs potentially increasing testosterone levels through enhanced cAMP production compared to ILCs. Protein-protein interaction analysis highlighted hub genes in cAMP signaling pathway, providing insights into molecular mechanisms of LCs regulating spermatogenesis.

Nlrp5 and Tle6 expression patterns in buffalo oocytes and pre-implantation embryos

Summary: This study aimed to investigate the expression of Nlrp5 and Tle6 genes in buffalo. The coding sequences of these two genes were successfully cloned and characterized. Results showed an absence of Nlrp5 or Tle6 transcripts in somatic tissues, except in the ovary. The expression levels of Nlrp5 and Tle6 in oocytes increased during early embryonic development and then decreased. Protein expression patterns were confirmed by immunofluorescence analysis.

Summary: This study conducted an identification, evolution, and expression analysis of the BRD gene family in the swamp buffalo. The BRD proteins were classified into six groups, and the BRD genes were irregularly distributed in 13 chromosomes. Comparative transcriptomics revealed differential expression of BRD genes in Sertoli cells at different developmental stages of buffalo.

Comparative Proteomic Analyses of Poorly Motile Swamp Buffalo Spermatozoa Reveal Low Energy Metabolism and Deficiencies in Motility-Related Proteins

Runfeng Liu, Xingchen Huang, Qinqiang Sun, Zhen Hou, Weihan Yang, Junjun Zhang, Pengfei Zhang, Liangfeng Huang, Yangqing Lu, Qiang Fu

Summary: Proteomic analysis of buffalo sperm reveals molecular mechanisms associated with poor motility, including inefficient energy metabolism, decreased sperm protamine proteins, a lack of motility-related proteins, and changes in tail structural proteins. These findings provide clues for the identification of molecular markers for poor motility in buffalo sperm.

Single-cell RNA sequencing uncovers dynamic roadmap and cell-cell communication during buffalo spermatogenesis

Liangfeng Huang, Junjun Zhang, Pengfei Zhang, Xingchen Huang, Weihan Yang, Runfeng Liu, Qinqiang Sun, Yangqing Lu, Ming Zhang, Qiang Fu

Summary: This study analyzed over 32,000 cells from buffalo testes using single-cell RNA sequencing, generating dynamic gene expression roadmaps of germ and somatic cell development. It uncovered the global cell-cell communication essential for regular spermatogenesis in the buffalo testicular microenvironment. These findings provide the theoretical basis for establishing buffalo germline stem cells in vitro or culturing organoids, as well as facilitating the expansion of superior livestock breeding.
