Summary: This study explored the role of CEA family member 5 (CEACAM5) in head and neck squamous cell carcinoma (HNSCC). The results showed a close association between CEACAM5 levels and lymph node (LN) metastasis, and high CEACAM5 levels had a positive impact on clinical prognosis. Experimental findings demonstrated that CEACAM5 inhibited cell proliferation and migration, and promoted apoptosis to suppress LN metastasis. Therefore, CEACAM5 has potential as both a prognostic marker and a therapeutic target in HNSCC.

Summary: The study revealed that the FGFR/RACK1 complex regulates senescence in LSCC cells by promoting the degradation of p53 through the formation of a complex with MDM2. Inhibition of FGFR or knockdown of RACK1 induced cell senescence, while inhibitors targeting MDM2 and p53 cooperatively inhibited colony formation and metastasis of LSCC cells. This understanding could have important implications for the treatment of LSCC.

Summary: Specific C-terminal nucleophosmin (NPM) mutations are associated with acute myeloid leukaemia and cause mislocalization of NPMmut to the cytoplasm. This leads to mislocalization of NPM-interacting partners, including the tumor suppressor p53. We discovered the formation of a ternary complex consisting of NPMmut, p53, and Mdm2, with p53 acting as a bridging link between Mdm2 and NPMmut. Silencing of p53 prevented mislocalization of Mdm2 in the presence of NPMmut.

Summary: TP53 mutation is a common genetic alteration in head and neck squamous cell carcinoma (HNSCC), and targeting p53 for therapy shows potential. Therapeutic strategies can be categorized into targeting wild-type p53 degradation or inhibition, directly affecting mutated p53, and specifically targeting HPV-positive HNSCC. This review discusses p53 regulation and its targeting in combination with existing therapies, based on the classification of HNSCC according to p53 mutation status and HPV infection.

Summary: Oral squamous cell carcinoma (OSCC) is the majority of oral cancers and the eighth most common cancer in men. Current chemotherapeutics have significant side effects, necessitating the development of new anti-cancer agents. In this study, new 1,2,3-triazole-chalcone hybrids were synthesized and evaluated against OSCC, showing great cytotoxicity and selectivity. The compound 1f was found to induce apoptosis and cell cycle arrest, and it also showed potential affinity for the MDM2 protein. Overall, 1f is a promising lead for a new chemotherapeutic drug against OSCC and possibly other types of cancers.

Summary: Inactivating and missense variants in the centrosomal CEP78 gene have been identified in CRDHL, along with a complex structural variant implicating CEP78. By using various sequencing and PCR techniques, researchers have characterized different SVs affecting CEP78 in individuals with CRDHL, demonstrating the importance of SV analysis in understanding the genetic basis of this rare retinal disease. Additionally, molecular consequences of these SVs on CEP78 expression were assessed, revealing potential mechanisms underlying CRDHL and highlighting the power of genome sequencing technologies in studying complex SVs in ocular diseases.
