The cathepsins belong to the enzymes of cathepsins family, which collect lysosomal proteinases. They have a wide spectrum of function in a lot of tissues and cell types. In pigs, they are extensively tested in relation to muscle transformation during dry-cured ham processing. Porcine CTSZ gene was mapped on SSC17 in the region which is suggestive of QTLs for meat quality. The CTSF and CTSD genes were localized on SSC2, at a distance of 13 cM. In the CTSZ and CTSF genes missense mutations g.37A > G and g.22G > C were identified. While in CTSD gene, SNP in 3'UTR was discovered (A70G). We investigated the associations between these three SNPs and economically important traits in Polish pigs. The significant effect of A70G CTSD polymorphism was detected for two growth traits. Animals carrying A/G genotype had lower daily gain and higher number days in test than both homozygotes (AG-853 g/day and 180 days; P < 0.01). The pigs with AA genotype of CTSD had higher level of meat traits and lower backfat thickness than GG pigs. Investigation of impact of mutation in CTSF on pig traits did not revealed obvious results. Analysis of mutation in CTSZ gene delivered two significant differences between GG and AG pigs, which were 75 g/day for TDG and 10 days NDT. To summarize, research on CTSD and CTSZ genes should be continued in order to find an additional QTL on SSC2 and SSC17 associated with economically important traits. (C) 2012 Elsevier B.V. Changes of Androstenone Concentrations in Saliva of Boars with Age

Kamila Pokorna, Jaroslav Citek, Petr Dolezal, Martyna Malopolska, Miroslav Tyra, Monika Okrouhla, Katerina Zadinova, Michal Sprysl, Nicole Lebedova, Roman Stupka

Summary: This study focused on finding a non-invasive method to determine boar taint in pigs by evaluating the concentration of androstenone compounds in their saliva. The results showed a correlation between the concentration of salivary androstenone and the age of the boars. This research has significant implications for preventing the occurrence of boar taint in pork.

Summary: The extracellular matrix (ECM) plays a crucial role in tissue structure and biochemical reactions, and dysregulation of the ECM can lead to diseases such as cancer. This study investigated the expression profiles of genes related to ECM remodeling and cell adhesion in equine sarcoid, the most common skin cancer in horses. The results suggest that abnormal expression of ECM-related genes is correlated with sarcoid formation. Understanding the molecular pathways involved in ECM remodeling and cell adhesion could contribute to the development of biomarkers and potential therapeutic targets for sarcoid-related cancer.

Variations in Fibrinogen-like 1 (FGL1) Gene Locus as a Genetic Marker Related to Fat Deposition Based on Pig Model and Liver RNA-Seq Data

Katarzyna Piorkowska, Kacper Zukowski, Katarzyna Ropka-Molik, Miroslaw Tyra

Summary: The goal of this study was to evaluate the effects of mutations in the FGL1 gene associated with pig productive traits. More than 50 crucial mutations were identified in the FGL1 gene. The association study showed significant effects of the FGL1 on various traits related to fat deposition. The study supports the theory that liver FGL1 is involved in the fat deposition process and provides a potential genetic marker for pig selection.

Summary: In this study, we investigated the genetic composition of the Polish draft horse (PDH) breed by analyzing whole mitochondrial d-loop variabilities and comparing them to sequences of other cold-blooded breeds. Our results showed high nucleotide diversity among the PDH population and the existence of two main haplogroups with strong kinship to the Belgian breed.

Summary: Chronic pancreatitis (CP) is an irreversible and progressive inflammatory disease. This study aimed to investigate the changes in pro-inflammatory cytokines, antioxidant defense system, and selected serum biochemical parameters in a cerulein-induced CP model in pigs. The study found that repetitive cerulein injections not only led to CP and induced inflammation in the intestine, but also impaired the intestinal barrier.

Influence of Media Composition on the Level of Bovine Satellite Cell Proliferation

Karolina Zygmunt, Agnieszka Otwinowska-Mindur, Katarzyna Piorkowska, Wojciech Witarski

Summary: It is predicted that by 2040, 35% of meat requirements will be provided through in vitro production. This study focused on developing a medium composition that effectively stimulates the proliferation of bovine satellite cells (BSCs) for cultured meat production. The results indicated that bovine serum and the addition of 10 ng/mL bFGF significantly enhanced the proliferation rate of BSCs, while low glucose content promoted the expression of crucial genes involved in satellite cell self-renewal.

The Effect of BSCL2 Gene on Fat Deposition Traits in Pigs

Katarzyna Piorkowska, Julia Sroka, Kacper Zukowski, Karolina Zygmunt, Katarzyna Ropka-Molik, Miroslaw Tyra

Summary: This study evaluated the effects of BSCL2 variants on slaughter and fattening characteristics and meat quality traits. The selection of mutations was based on variant calling analysis and chi test results using subcutaneous fat RNA-seq data. Significant genotype/allele distribution variations between high- and low-fat pigs were observed. The study suggests that variant calling from RNA-seq data may help develop genetic markers for complex pig traits, and the BSCL2 G allele (rs341493267) was found to increase backfat thickness in pigs.

Equine Metabolic Syndrome: A Complex Disease Influenced by Multifactorial Genetic Factors

Monika Stefaniuk-Szmukier, Katarzyna Piorkowska, Katarzyna Ropka-Molik

Summary: Equine metabolic syndrome (EMS) is an important issue in modern veterinary medicine, linked to the painful hoof laminitis. We have summarized available information on the genetic background, environmental factors, diagnosis, treatment, and management of EMS for both scientific and practical purposes. Clinical presentation in horses includes obesity, lameness, and insulin dysregulation, with ongoing research in regenerative medicine for promising therapies.

New long-non coding RNAs related to fat deposition based on pig model

Katarzyna Piorkowska, Kacper Zukowski, Katarzyna Ropka-Molik, Miroslaw Tyra

Summary: This study used pigs as an animal model to investigate the potential gene expression regulation by lncRNAs in fat tissue. The results showed that the expression of hundreds of genes, including 8 lncRNAs, was dependent on fat content. It was also discovered that a transcript related to human insulin resistance was identified, and some lncRNAs may play a regulatory role in gene expression.
