Summary: POLE2 is significantly upregulated in lymphoma tissues and cells. Knockdown of POLE2 inhibits proliferation and migration, induces apoptosis and cycle arrest in lymphoma cells, and impairs tumor growth in mice. POLE2 may serve as a novel therapeutic target for lymphoma by inhibiting Wnt/β-catenin signaling pathway.

Summary: Classical Hodgkin lymphoma is a relatively common lymphoma that is generally considered highly curable with standard first-line chemotherapy and radiotherapy, although some patients may not be cured or may experience late toxic effects leading to premature death. Recent changes in the gold standard of chemotherapy, now often including immunotherapy in relapse settings, have further impacted treatment outcomes.

Summary: CK1 delta/epsilon enhance beta-catenin-mediated transcription by regulating beta-catenin acetylation. CK1 delta/epsilon interact with Tip60 and facilitate its recruitment to the beta-catenin complex, leading to increased beta-catenin acetylation at K49. Furthermore, this study discovers that a complex consisting of CK1 delta/CK1 epsilon/beta-catenin/Tip60 is present in colon cancer cells, and this complex plays an important role in beta-catenin acetylation.

Summary: Endometriosis, common in women of childbearing age, involves the presence of endometrial tissue outside the uterine cavity. Research shows that WEE1 promotes migration and fibrosis in endometrial stromal cells through the Wnt/β-catenin signaling pathway, suggesting it may be a potential therapeutic target for endometriosis treatment.

Summary: Colorectal cancer is a major cause of cancer-related deaths worldwide, with APC gene mutations playing a key role in tumor development. The novel cytotoxin MakA induces apoptosis in cancer cells, with potent cytotoxic activity against cancer cell lines while sparing nontransformed cells. MakA inhibits beta-catenin-mediated tumor cell proliferation, making it a promising candidate for therapeutic development against colon cancer.

Summary: SOX9 promotes turnover of β-catenin independently of the destruction complex and proteasome in mammalian cell culture. This requires the activation of MAML2, which acts as a coactivator for SOX9 in antagonizing the Wnt/β-catenin pathway. This newly identified pathway provides a parallel mechanism for restricting Wnt/β-catenin signaling.

Summary: Osteoarthritis is a chronic disease that lacks disease-modifying treatment, with the Wnt/beta-catenin pathway playing a key role in its pathophysiology. Both excessive stimulation and suppression of this pathway can contribute to the development of OA. MicroRNAs have been found to regulate various cellular processes in different diseases, offering potential insights for the development of miRNA-based therapeutics for OA.
