Differential expression of NF-κB target genes in MALT lymphoma with and without chromosome translocation: insights into molecular mechanism

Mucosa-associated lymphoid tissue (MALT) lymphoma is characterized by t(11;18)(q21;q21)/API2-MALT1, t(1;14)(p22;q32)/BCL10-IGH and t(14;18)(q32;q21)/IGH-MALT1, which commonly activate the nuclear factor (NF)-kappa B pathway. Gastric MALT lymphomas harboring such translocations usually do not respond to Helicobacter pylori eradication, while most of those without translocation can be cured by antibiotics. To understand the molecular mechanism of these different MALT lymphoma subgroups, we performed gene expression profiling analysis of 21 MALT lymphomas (13 translocation-positive, 8 translocation-negative). Gene set enrichment analysis (GSEA) of the NF-kappa B target genes and 4394 additional gene sets covering various cellular pathways, biological processes and molecular functions have shown that translocation-positive MALT lymphomas are characterized by an enhanced expression of NF-kappa B target genes, particularly toll like receptor (TLR)6, chemokine, CC motif, receptor (CCR)2, cluster of differentiation (CD) 69 and B-cell CLL/lymphoma (BCL)2, while translocation-negative cases were featured by active inflammatory and immune responses, such as interleukin-8, CD86, CD28 and inducible T-cell costimulator (ICOS). Separate analyses of the genes differentially expressed between translocation-positive and -negative cases and measurement of gene ontology term in these differentially expressed genes by hypergeometric test reinforced the above findings by GSEA. Finally, expression of TLR6, in the presence of TLR2, enhanced both API2-MALT1 and BCL10-mediated NF-kappa B activation in vitro. Our findings provide novel insights into the molecular mechanism of MALT lymphomas with and without translocation, potentially explaining their different clinical behaviors. Summary: Colorectal cancer is a deadly disease with increasing rates worldwide. Caldesmon (CaD) has emerged as a promising biomarker for diagnosis and therapy. It was found that l-CaD is overexpressed in colorectal cancer and associated with preinvasive stages, while h-CaD is expressed in smooth muscle cells but not in cancer cells or normal colon mucosal epithelial cells. These findings have significant implications for the management of colorectal cancer patients.

Summary: Distinguishing FL from NMZL can be difficult in cases with unusual morphologic and phenotypic features and without characteristic cytogenetic rearrangements. Ancillary techniques, such as FISH, RT-MLPA, and NGS, can provide diagnostic contributions for unclassified tumors. In this study, FISH detected 1p36 deletion in some FLs and unclassified tumors, while RT-MLPA signatures were associated with FL and NMZL subtypes. Combining RT-MLPA and NGS findings successfully discriminated most unclassified tumors towards FL or NMZL diagnosis.

The Role of Vitamin D Supplementation on Airway Remodeling in Asthma: A Systematic Review

Summary: Asthma, a common chronic respiratory disease affecting millions of people worldwide, is being increasingly linked to vitamin D deficiency. This systematic review evaluated the effects of vitamin D supplementation on airway remodeling in asthmatic patients. After a thorough search of several databases, 9 experimental studies were included, suggesting that vitamin D inhibits airway smooth muscle cell contraction and remodeling, reduces inflammation, regulates collagen synthesis in the airways, and modulates the action of bronchial fibroblasts. However, the role of TGF-beta 1 in impairing vitamin D-induced airway epithelial host defense mechanisms needs further investigation.

Insights into the Molecular Mechanisms Mediating Extravasation in Brain Metastasis of Breast Cancer, Melanoma, and Lung Cancer

In this review, the molecular events that occur during extravasation in breast cancer, melanoma, and lung cancer, which are most likely to develop brain metastasis, are summarized and compared. This review provides valuable information for the development of therapeutic strategies for the prevention or treatment of brain metastases.

The case review emphasized the challenges in diagnosing marginal zone lymphomas, including distinguishing between different types and the need for molecular studies to establish a final diagnosis. The workshop also addressed the overlapping spectrum between pediatric nodal marginal zone lymphoma and pediatric-type follicular lymphoma.

Titanium Particles Modulate Lymphocyte and Macrophage Polarization in Peri-Implant Gingival Tissues

Summary: This study analyzed the gene expression in peri-implant gingival tissues of patients with failed, successful, or no implants. The results showed that titanium particles modulate lymphocyte and macrophage polarization in peri-implant tissues, which may contribute to implant failure. The presence of titanium particles and infiltration of immune cells, particularly lymphocytes and macrophages, were confirmed in the failed implant samples. In vitro validation also demonstrated increased expression of IL-1 beta, IL-8, and IL-18 by macrophages.

Molecular Heterogeneity of the Brain Endothelium

Summary: The blood-brain barrier (BBB) is a neurovascular structure in the brain that maintains brain homeostasis but hinders drug delivery. Despite advances in understanding the BBB, efficiently treating brain diseases by overcoming the BBB remains challenging. Gene and protein expression analysis of the BBB in different brain regions showed variation, indicating the importance of considering the BBB heterogeneity for effective drug delivery and brain disease treatment in future research programs.

Mutational Profile and Pathological Features of a Case of Interleukin-10 and RGS1-Positive Spindle Cell Variant Diffuse Large B-Cell Lymphoma

Summary: We report a rare variant of diffuse large B-cell lymphoma with spindle cell morphology in a 74-year-old male patient who presented with right supraclavicular (lymph) node enlargement. The lymphoma was characterized by a cell-of-origin subtype of germinal center B-cell-like (GCB), and mutational profiling confirmed mutations in multiple genes associated with aggressive B-cell lymphomas. The immune microenvironment showed infiltration of tumor-associated macrophages and expression of certain markers associated with poor prognosis. The patient achieved a complete response with R-CHOP therapy.
