PRL3 enhances T-cell acute lymphoblastic leukemia growth through suppressing T-cell signaling pathways and apoptosis

E. G. Garcia, A. Veloso, M. L. Oliveira, J. R. Allen, S. Loontiens, D. Brunson, D. Do, C. Yan, R. Morris, S. Iyer, S. P. Garcia, N. Iftimia, W. Van Loocke, F. Matthijssens, K. McCarthy, J. T. Barata, F. Speleman, T. Taghon, A. Gutierrez, P. Van Vlierberghe, W. Haas, J. S. Blackburn, D. M. Langenau

Summary: The study identifies PRL3 as a collaborating oncogenic driver in T-ALL, which synergizes with MYC to promote leukemia development and suppress apoptosis in cells. By suppressing downstream phosphorylation signaling pathways, PRL3's role in T-ALL is elucidated.

Immunophenotype of Gastric Tumors Unveils a Pleiotropic Role of Regulatory T Cells in Tumor Development

Sara Rocha, Afonso P. Basto, Marieke E. Ijsselsteijn, Sara P. Teles, Maria M. Azevedo, Gilza Goncalves, Irene Gullo, Gabriela M. Almeida, Joaquin J. Maqueda, Marta Oliveira, Fatima Carneiro, Joao T. Barata, Luis Graca, Noel F. C. C. de Miranda, Joana Carvalho, Carla Oliveira

Summary: The role of regulatory T cells (Tregs) in gastric cancer (GC) remains controversial. Tregs may have a non-immunosuppressive effect on intestinal-type GC by inducing membrane expression of IL2R alpha and activating the MAPK signaling pathway, promoting spheroid growth in early stages of tumor development. This finding could lead to novel therapeutic strategies for intestinal-type GC and other tumors with similar immune context.

Overexpression of wild-type IL-7Rα promotes T-cell acute lymphoblastic leukemia/lymphoma

Ana Silva, Afonso R. M. Almeida, Ana Cachucho, Joao L. Neto, Scifie Demeyer, Mafalda de Matos, Thea Hogan, Yunlei Li, Jules Meijerink, Jan Cools, Ana Rita Grosso, Benedict Seddon, Joao T. Barata

Summary: Overexpression of IL-7R alpha can promote T-cell tumorigenesis and lead to malignancies in various organs, involving activation of multiple signaling pathways and sensitivity to inhibitors of IL-7R-mediated signaling. Established tumors may no longer require high levels of IL-7R expression upon secondary transplantation, highlighting potential therapeutic implications.

Summary: This study reveals that activating mutations in Interleukin-7 receptor alpha can initiate preleukemia in primary human hematopoietic progenitors and cooperate with CDKN2A silencing in the progression into acute lymphoblastic leukemia (BCP-ALL).
