Summary: This study investigated the protein expression of CDK7 in NSCLC and found that CDK7 is overexpressed in squamous cell carcinomas (sqNSCLC). High expression of CDK7 is associated with lymph node metastases and a poor prognosis in sqNSCLC. CDK7 may serve as a novel prognostic biomarker and a potential target for future anti-cancer therapy in NSCLC.

Summary: In this study, the expression and function of Lnc-LSAMP-1 in NSCLC were investigated. It was found that Lnc-LSAMP-1 was significantly down-regulated in NSCLC tissues and its loss expression was associated with unfavorable prognosis. Over-expression of Lnc-LSAMP-1 inhibited lung cancer cell proliferation, invasion, and migration, and enhanced chemotherapy sensitivity. Mechanistically, Lnc-LSAMP-1 formed a complex with LSAMP gene, protecting it from degradation.

Summary: Lung cancer is the leading cause of cancer-related deaths globally, with most patients still being diagnosed at an advanced stage despite screening efforts for high-risk populations. In the past decade, management of lung cancer has significantly progressed towards personalized treatments, with molecular biomarkers playing a crucial role in prognosis and predicting therapy response. Non-small cell lung cancer, being the most common type, is the primary focus for discussion on promising new biomarkers in clinical management.

Summary: This study confirms the specific localization of VDR in gastric cancer tissues and demonstrates that activation of VDR can suppress CAF-induced chemotherapy resistance in gastric cancer. The study also reveals that CAF-derived IL-8 induces oxaliplatin resistance through activating the PI3K/AKT pathway, and treatment with vitamin D can attenuate the tumor-supportive effect of IL-8 on gastric cancer cells.

Summary: Despite progress in cancer treatment, gastric cancer (GC) still has a poor prognosis. Recent research has shown that the malignant behavior of cancer is not only caused by tumor cells, but also mediated by the activity of cancer stroma and regulated by molecular networks in the tumor microenvironment (TME). Cancer-associated fibroblasts (CAFs) are abundant in the stroma and play a significant role in the malignant development of GC. CAFs can modulate the biological properties of tumor cells through various mechanisms, affecting the initiation and progression of GC. However, there is heterogeneity in the cellular origins, phenotypes, and functions of CAFs in the TME of GC, and the molecular mechanisms are not well understood.

Summary: PROX1 is a transcription factor that promotes malignant transformation and stemness in colorectal cancer (CRC). It has been found that PROX1 is associated with cell glucose metabolism and regulates cell proliferation and glucose metabolism through interaction with SIRT3. High PROX1 expression combined with low SIRT3 expression predicts poor prognosis in CRC patients.

Summary: In this study, the researchers identified a relationship between branched-chain amino acid metabolism and cell growth and motility through RhoC signaling. They found that enhanced BCAT1 activity, caused by a mutation enriched in gastric cancer, leads to increased production of a metabolite that activates RhoC signaling. The study also found that the BCAA-derived metabolite can directly bind to the small GTPase protein RhoC and enhance its activity. The researchers also discovered that the drug candesartan can inhibit BCAT1 activity, leading to decreased RhoC activity and cancer cell motility.

Summary: The present study investigates the mechanism of gambogenic acid (GNA), a natural compound, on apoptosis and ferroptosis in colorectal cancer (CRC). The results show that GNA inhibits tumor growth by suppressing the proliferation of CRC cell lines and induces ferroptosis by activating endoplasmic reticulum stress and ROS production. Furthermore, GNA regulates iron metabolism to further induce ferroptosis.

Summary: This study investigated the immune microenvironment of cutaneous neurofibromas (cNFs) in patients with neurofibromatosis 1 (NF1). The results showed that cNFs have substantial populations of T cells and macrophages, which may be tumor-specific. T cell populations in cNFs were found to be different from those in the skin, and cNFs exhibited lower expression of proteins related to T cell-mediated immunity compared to the skin.
