Summary: We present a large-scale carboxylation of N-Boc-4,4-difluoropiperidine (1) enabled by a continuous flow process. The flow process involved N-Boc-directed alpha-deprotonation using s-BuLi in THF and subsequent trapping with CO2 gas. Flow chemistry allowed for safe and scalable preparation of 400 g of carboxylic acid 2 in a day to support our medicinal chemistry research program.

Summary: In this study, the encapsulation of Gusperimus in PLGA-PEG nanoparticles was investigated to achieve controlled release and protect its stability and bioactivity. The effects of different surfactants and polymer concentrations on the properties of the nanoparticles were evaluated. The results showed that the system prepared with 1.7% w/v PLGA-PEG and 1% w/v Pluronic (R) F127 had the smallest particle size and higher colloidal stability. The nanoparticles exhibited high encapsulation efficiency and load capacity, as well as anti-inflammatory effects in macrophages.

Summary: In this study, the scaling up of toluene nitration in a meso-scale flow reactor was conducted with consideration of safety. The exothermic characteristic of the reaction was demonstrated and numerical simulation was used to study the hydrodynamics. The effects of flow rates, temperatures, and molar ratios on the reaction were analyzed, and a comparison between batch calorimeter and flow reactor performances was presented.

Summary: A droplet-based microreactor system was designed to overcome the low efficiency of mass and heat transfer in conventional batch synthesis of Ag NPs. By regulating residence time and introducing seed-mediated growth, continuous and controllable synthesis of Ag NPs with sizes ranging from 7 to 30 nm, polydispersity indexes less than 10%, and high stability were achieved. This system provides a diffusion-limited reaction model for the synthesis of metal nanomaterials.

Summary: This article presents a bottom-up nanowelding strategy for the fabrication of high-purity nanoporous gold (NPG) from gold nanoparticles (NPs). By transferring the layered Au NP film onto a solid substrate and conducting direct current (DC) nanowelding, NPG can be produced at low temperatures within 10 seconds without damaging the spherical structure. This method also allows for the tunability of thickness, ligament size, and pore size by using differently sized Au NPs as starting building blocks, providing great flexibility for creating functional porous nanomaterials.

Summary: This study proposes the use of polyethylene glycol 400 (PEG) and trehalose as a surface modification technique to improve PLGA-based nanoparticles as drug carriers for neurons. The combination of PEG and trehalose enhances the hydrophilicity of the nanoparticles and promotes their cellular internalization. Through optimization using central composite design, PLGA nanoparticles with diameters smaller than 200 nm were produced, and the coating process did not significantly increase their size. The nanoparticles showed high curcumin entrapment efficiency and a controlled release profile.

Summary: In this study, the post-processing methods of the stereolithography (SLA) printing method were compared to assess their effects on the surface properties. It was found that polishing could improve transparency, although achieving glass-like transparency was challenging. The use of a commercially available vehicle scratch remover or silicone oil could effectively enhance transparency. A method combining sequential grinding with a scratch remover was proposed for achieving high transparency. However, it was difficult to achieve a contact angle of 90 degrees or more, even with various surface treatment methods.
