Summary: This study developed a comprehensive histopathologic grading scheme for murine knee osteoarthritis (OA) that covers all-stage osteoarthritic changes in all major joint tissues. The new grading scheme showed high reproducibility and accuracy in assessing OA severity and progression. It is the first whole-joint grading scheme that includes periarticular soft-tissue ossification and is suitable for quick OA severity assessments of the entire femorotibial joint.

Summary: There are currently no known drugs or treatment methods to prevent post-traumatic osteoarthritis (PTOA), but cationic delivery carriers can help overcome the challenge of drug delivery. In this study, a cationic green fluorescent protein (GFP) was used as a carrier to deliver Insulin-Like Growth Factor 1 (IGF-1) for potential therapeutic treatment of PTOA. Through in vitro experiments and the development of a predictive mathematical transport model, the study provides a foundation for translating these findings into preclinical and potentially clinical applications.
