Familial co-segregation and the emerging role of long-read sequencing to re-classify variants of uncertain significance in inherited retinal diseases

Summary: Phasing genetic variants is crucial for identifying potential disease-causing variants. Reclassification of variants of uncertain significance (VUS) in autosomal recessive inherited retinal diseases (IRDs) can lead to a genetic diagnosis when combined with familial co-segregation analysis. This study reports four cases where a VUS was found to be trans to a known pathogenic variant through familial co-segregation, resulting in the reclassification of the VUS to likely pathogenic and a genetic diagnosis. Additionally, targeted long-read sequencing was utilized to infer variant phasing in a simplex patient without access to family members for co-segregation analysis. This emerging method provides a solution for cases where genetic testing of family members is unfeasible, allowing for a complete genetic diagnosis.

Novel homozygous variant in ARL2BP associated with retinitis pigmentosa, situs inversus, and male infertility in a Chinese patient

Summary: We identified a Chinese patient carrying a novel ARL2BP variant, who presented with retinitis pigmentosa, situs inversus, and oligozoospermia. This is the first report of the coexistence of situs inversus and male infertility in ARL2BP variant patients. Additionally, the patient likely had olfactory dysfunction.

Beyond nephronophthisis: Retinal dystrophy in the absence of kidney dysfunction in childhood expands the clinical spectrum of CEP83 deficiency

Summary: CEP83 protein plays a crucial role in ciliogenesis and deficiency can lead to ciliopathy. Two unrelated patients with retinitis pigentosa were found to have bi-allelic variants in the CEP83 gene, without nephronophthisis or kidney dysfunction. This study expands the clinical spectrum of CEP83 deficiency and suggests including CEP83 in gene panels for inherited retinal diseases for timely diagnosis.

Rare Co-Occurrence of Visual Snow in a Female Carrier With RPGRORF15-Associated Retinal Disorder

Summary: XLRP, primarily caused by mutations in the RPGR gene, manifests as severe phenotypes and early disease onset in affected males, while female carriers are often asymptomatic or exhibit stable phenotypes. A case study reported an 8-year-old female carrier who experienced early progressive cone-rod pattern retinal dystrophy and visual snow-like symptoms, despite harboring a heterozygous pathogenic RPGR variant and random X-inactivation pattern. Phenotypic variations were observed within the family despite sharing an identical RPGR variant with the father. Information on genotypic and phenotypic diversity among XLRP carriers is crucial for diagnosis, management, and monitoring, as well as the development of gene therapy trials.

Summary: Usher syndrome is a genetically heterogeneous disease with highly variable clinical manifestations. A recent study has found that variants in the ARSG gene are responsible for Usher syndrome type IV, expanding the phenotypic classification of the disease. This study describes the phenotype of three unrelated individuals with Usher syndrome type IV, identifying multiple novel pathogenic variants and confirming the loss of sulfatase activity in the mutant proteins through functional experiments.

Is RPGR-related retinal dystrophy associated with systemic disease? A case series

Two cases co-presented with confirmed diagnoses of primary ciliary dyskinesia and one case with multiple sino-respiratory symptoms strongly suggestive of primary ciliary dyskinesia. These and all previously reported RPGR co-pathologies relate to ciliopathies and have no other systemic associations.

X-linked dominant RPGR gene mutation in a familial Coats angiomatosis

The Authors suggest RPGR gene screening mutations in patients presenting this phenotype.

Molecular pathology of Usher 1B patient-derived retinal organoids at single cell resolution

Summary: This study investigated the pathomechanism of Usher syndrome type 1B (RP) using 3D retinal organoids generated from induced pluripotent stem cells derived from USH1B patients. The results revealed differential gene expression related to mitochondrial functions, proteasomal protein degradation, and RNA splicing. Single-cell RNA sequencing showed MYO7A expression in rod photoreceptor and Mu center dot ller glial cells, suggesting involvement in stress responses and apoptotic signaling pathways. This human model provides important insights for the development of therapeutic strategies.

A Genotype-Phenotype Analysis of the Bardet-Biedl Syndrome in Puerto Rico

Further research should focus on understanding the impact of the c.1645G>T (p.Glu549*) mutation on the BBS1 gene and protein product.

Development and testing of an artificial intelligence tool for predicting end-stage kidney disease in patients with immunoglobulin A nephropathy

Neurocognitive assessment and DNA sequencing expand the phenotype and genotype spectrum of Alstrom syndrome

The research found that patients with a milder phenotype performed better on certain neurocognitive tests compared to those with a typical phenotype, and deficits in auditory working memory and apraxia were common among all patients.

Paternal Uniparental Isodisomy of Chromosome 2 in a Patient with CNGA3-Associated Autosomal Recessive Achromatopsia

Summary: This study identified a homozygous variant in the CNGA3 gene in a patient with ACHM, with the variant being detected in a heterozygous state in the father but not in the mother. The homozygosity of the CNGA3 variant in the patient was found to be due to partial or complete paternal uniparental isodisomy. In addition to the ACHM phenotype, the patient was clinically unsuspicious and healthy.

Characterization of Human Subcutaneous Adipose Tissue and Validation of the Banking Procedure for Autologous Transplantation

Summary: This study aimed to characterize and validate the storage effectiveness of cryopreserved adipose tissue (AT) for up to three years for clinical applications. AT components were isolated from fresh and cryopreserved samples and cell viability was assessed. Adipogenic and endothelial precursors and mature cells were measured. The differentiation potential and cellular composition of cryopreserved AT were maintained. Two patients successfully underwent autologous cryopreserved AT transplants without complications.

Obesity, the Adipose Organ and Cancer in Humans: Association or Causation?

Summary: Epidemiological observations, experimental studies and clinical data suggest that obesity is associated with an increased risk of developing various types of cancer. The adipose tissue alterations occurring in obesity could play a significant role in this association. The obesity-altered adipose tissue environment contributes to cancer development, progression, metastasis and chemoresistance through direct and indirect interactions involving different cellular types and molecular mechanisms.
