Summary: This article discusses the clinical significance of Klotho and its potential as a biomarker in chronic kidney disease (CKD). However, there is inconsistent and contradictory evidence regarding the relationship between Klotho and CKD, and a consensus has not been reached. The authors systematically assess the areas of controversy and highlight future directions to resolve inconsistencies and clarify the role of Klotho in CKD clinical practice.

Summary: The study found that CD patients have more M2 monocytes and a disturbed cytokine balance. This suggests that these immunological markers could serve as specific diagnostic biomarkers for CD.

Summary: Autosomal-Dominant Polycystic Kidney Disease (ADPKD) is a genetic disorder caused by mutations in PKD1 or PKD2 genes. The involvement of autophagy in ADPKD remains a subject of investigation, with potential implications on cyst formation and fibrosis. Autophagy inducers have shown promising results in preclinical studies and may provide a potential avenue for future investigations.

Summary: PAFAH1B3 expression is significantly increased in HCC patients, and high expression is associated with poorer overall survival and disease-free survival. PAFAH1B3 may be involved in HCC through cell cycle, cell metabolism, spliceosome, and RNA transport. Inhibiting PAFAH1B3 impairs HCC cell proliferation, invasion, and migration, while downregulating glycolysis and lipid synthesis pathways.

Summary: Serum serotonin levels were found to be decreased in COVID-19 patients with more severe disease, along with increased kynurenine and decreased tryptophan concentrations. Lower serotonin concentrations were associated with subsequent worsening of moderate disease, suggesting it may be a valuable biomarker for disease severity and prognosis.

Summary: In this study, the researchers investigated whether DKK3 could improve outcome prediction in ADPKD. The results demonstrated that ADPKD patients had significantly higher uDKK3 levels, and uDKK3 correlated positively with the Mayo classification and kidney function. Although the effect was no longer significant in multivariate regression analysis adjusting for other factors, adding copeptin levels and the interaction term between copeptin and uDKK3 to the model improved the variance explanation of eGFR slope. Future studies are needed to confirm these findings, but uDKK3 appears to be an attractive candidate for improving outcome prediction in ADPKD.

Summary: New image-derived biomarkers are needed to improve the management of autosomal dominant polycystic kidney disease. A new technique to individually segment cysts and quantify biometric parameters has been developed. Total cyst number and cyst parenchyma surface area showed superior prediction of kidney function decline and disease stages compared to the measurement of total kidney volume.

Summary: High expression of ASXL2 in pancreatic cancer is associated with poor overall survival and is mainly involved in inflammatory response and epithelial mesenchymal transition. Patients with high ASXL2 expression are more likely to benefit from certain treatment modalities such as immune checkpoint blockade and specific chemotherapeutic drugs.

Summary: Using CRISPR/Cas9 mutagenesis, researchers established models for skeletal ciliopathies in Xenopus tropicalis and identified Ttc30a as a key gene related to known skeletal ciliopathy genes, with loss of Ttc30a leading to limb deformities and renal cysts. The study revealed TTC30A/B as essential nodes in the network of ciliary chondrodysplasia and nephronophthisis-like disease proteins.

Summary: PIK3C2A expression is decreased in various tumors, particularly in renal clear cell carcinoma (KIRC). It is significantly correlated with tumor stage, grade, and other clinicopathological features in KIRC. PIK3C2A serves as a potential biomarker for tumor progression and is associated with immune infiltration in KIRC.

Summary: Our study revealed that LRRK2 plays a crucial role in the tumorigenesis and progression of ccRCC, and may serve as a potential predictor and therapeutic target for this type of cancer. The up-regulation of LRRK2 in ccRCC was found to be correlated with DNA methylation and associated with a better prognosis in patients with ccRCC.

Summary: This review discusses the efficacy and safety of dietary interventions, such as caloric restriction, intermittent fasting, and ketogenic diet, in managing ADPKD. These interventions show promise in improving metabolic health and reducing oxidative stress in ADPKD, but limited clinical evidence supports their effectiveness.
