An experiment was performed to examine the effect of dietary copper supplementation on weight gain, neuropeptide Y (NPY) concentration and NPY mRNA expression level in the hypothalamus of pigs. Forty-five crossbred pigs were randomly assigned to three groups of 15 pigs, each comprising five replicates of 3 animals. Pigs were allocated to diets that contained 10mg/kg (as a control), 125 and 250mg/kg copper as CuSO4. Live weight gain and feed conversion efficiency was determined at the end of the experiment and five pigs, selected at random from each group, were slaughtered and the hypothalami collected for determination of NPY concentration and NPY mRNA expression level. The results showed that average daily gain (ADG) and average daily feed intake (ADFI) were higher and feed:gain (F:G) ratio was lower in pigs fed the diets with 125 and 250 mg/kg copper (P<0.05), respectively, than in pigs fed a diet with 10 mg/kg copper, but that there was no statistically significant difference in growth performance between animals of the 125 mg/kg and the 250 mg/kg copper groups. Furthermore, pigs fed diets with 125 and 250 mg/kg copper had higher NPY concentrations and NPY mRNA expression levels in their hypothalamus than control animals. The data indicated that 125 and 250 mg/kg copper gave similar responses in terms of weight gain, whilst high dietary copper could enhance NPY concentration and NPY mRNA expression level in the hypothalamus of pigs. High dietary copper appears to increase feed intake and promote weight gain by enhancing NPY concentration and NPY mRNA expression level in the hypothalamus of pigs. (C) 2007 Elsevier GmbH. Salt-contaminated water inducing pulmonary hypertension and kidney damage by increasing Ang II concentration in broilers

Yuxuan Peng, Xiaoqi Yang, Hao Li, Mudassar Iqbal, Aoyun Li, Jiabin Zhang, Mengdi Zhang, Jiakui Li, Donghai Zhou

Summary: This study found that drinking low-salt and high-salt contaminated water can lead to increased ascites heart index and hematocrit in broilers, as well as significant increases in blood urea nitrogen and creatinine levels. The kidneys may show vacuolar and fibrotic degeneration, with clear signs of apoptosis. Furthermore, the study showed that the transcription levels of AGT, REN, and ACE genes related to the RAAS pathway increased significantly after using salt-contaminated water, along with a significant rise in the concentration of angiotensin II (Ang II). The research also demonstrated that exogenous Ang II treatment could decrease the cell activity and promote apoptosis in chicken embryonic kidney cells, suggesting that salt-contaminated water may exacerbate PHS and kidney damage through the increase of Ang II.

Summary: Tibial dyschondroplasia (TD) is a common metabolic disorder in broilers caused by thiram consumption. It impairs bony and cartilage processes, resulting in lameness and mortality in commercial poultry. Cellular pathways involving anti-apoptotic, pro-apoptotic, and executioner caspases regulate the chondrocyte changes. The inflammatory response mediated by inflammasomes and mitochondrial-mediated apoptosis are important pathways in TD.

The Effect of miR-140-5p with HDAC4 towards Growth and Differentiation Signaling of Chondrocytes in Thiram-Induced Tibial Dyschondroplasia

Wangyuan Yao, Muhammad Fakhar-e-Alam Kulyar, Yanmei Ding, Haitao Du, Jiajia Hong, Kyein San Loon, Shah Nawaz, Jiakui Li

Summary: The interaction between miR-140 and HDAC4 plays a crucial role in bone disorders such as tibial dyschondroplasia. Low miR-140 expression is associated with poor pathological characteristics and low chondrocyte survival. HDAC4 inhibits MEF2C and Runx2 and interacts with Col-II, Col-X, and COMP, leading to uncontrolled chondrocyte proliferation, differentiation, and dysplastic growth. Targeting the miR-140 and HDAC4 feedback loop may provide a promising therapeutic approach for chondrocyte-related bone disorders.
