Summary: This study identifies FAM83A as an oncogene of HNSCC with overexpression significantly associated with tumor size, lymph node status, and clinical tumor stages. FAM83A promotes HNSCC cell growth and metastasis by inducing EMT through activation of the Wnt/beta-catenin signaling pathway. Knockdown of FAM83A suppresses tumor growth and distant metastasis in xenograft animal models of HNSCC.

Summary: The study revealed that XRN2 is significantly overexpressed in oral squamous cell carcinoma (OSCC) and closely associated with tumor differentiation, prognosis, and metastasis. Functional assays in vitro showed that down-regulation of XRN2 inhibited cell proliferation, migration, and invasion while promoting apoptosis, and also affected the epithelial-mesenchymal transition (EMT) characteristics of OSCC cells.

Summary: This study found that phosphoglycerate dehydrogenase (PHGDH) was highly expressed in esophageal squamous carcinoma (ESCC), and high expression was associated with poor prognosis. In vitro and in vivo experiments demonstrated that PHGDH knockdown inhibited ESCC progression by suppressing Wnt/beta-catenin signaling pathway. These results indicate that PHGDH may be a potential therapeutic target for ESCC.

Summary: Esophageal cancer can metastasize to distant sites, leading to poor prognosis. Intestinal metastasis is extremely rare with atypical clinical presentations. We present a case of rectal metastasis after surgery for esophageal squamous cell carcinoma. The patient achieved good short-term efficacy with adjuvant chemoradiotherapy and carrelizumab immunotherapy, and is currently in tumor-free survival. This case report aims to deepen our understanding of rare metastasis in esophageal squamous cell carcinoma and promote the use of local radiotherapy, chemotherapy, and immunotherapy to improve survival.

Summary: This study investigated the expression of GPBAR1 in HCC tissues and tumor-adjacent tissues and found that GPBAR1 was highly expressed in HCC tissues. GPBAR1 expression was significantly associated with poor prognosis in HCC and served as an independent prognostic biomarker. Additionally, GPBAR1 expression was significantly correlated with asynchronous bone metastasis but not liver or lung metastases. These findings provide new insights into HCC metastasis and improve prognostic assessment of HCC.

Summary: The protein gelsolin plays a significant role in controlling cytoskeletal remodeling, cell morphology, differentiation, movement, and apoptosis. Its expression has been found to be decreased in various pathological conditions and it has emerged as a potential diagnostic biomarker for early disease detection. Gelsolin can function as either an oncoprotein or a tumor suppressor depending on the type of carcinoma. This review discusses the roles of gelsolin in inflammation and wound healing, cancers, and tumor microenvironment, as well as future prospects for related studies and clinical application.

Summary: RAB37 GTPase regulates cargo exocytosis by cycling between GDP-bound and GTP-bound forms. It also simultaneously regulates autophagy activation and TIMP1 secretion in lung cancer cells under starvation conditions. Secretory autophagy participates in TIMP1 secretion in a RAB37- and Sec22b-dependent manner. Autophagy induction facilitates TIMP1 secretion and suppresses the motility and metastasis of lung cancer cells in a RAB37-dependent manner in a lung-to-lung mouse model.

Summary: WDR4/PTPN23 axis plays a central role in the trafficking of EGFR and c-MET in non-small cell lung cancer (NSCLC). WDR4-mediated PTPN23 ubiquitination inhibits lysosome trafficking and degradation, sustaining EGFR and c-MET signaling and promoting NSCLC proliferation, migration, and metastasis. Clinical studies show that low expression of PTPN23 is associated with high expression of WDR4 and poor prognosis in lung cancer. Targeting WDR4-mediated PTPN23 ubiquitination may be a potential therapeutic strategy for EGFR TKI-resistant NSCLC.
