Identification of Putative ORF5 Protein of Porcine Circovirus Type 2 and Functional Analysis of GFP-Fused ORF5 Protein

Porcine circovirus type 2 (PCV2) is the essential infectious agent responsible for causing porcine circovirus-associated diseases in pigs. To date, eleven RNAs and five viral proteins of PCV2 have been detected. Here, we identified a novel viral gene within the PCV2 genome, termed ORF5, that exists at both the transcriptional and translational level during productive infection of PCV2 in porcine alveolar macrophages 3D4/2 (PAMs). Northern blot analysis was used to demonstrate that the ORF5 gene measures 180 bp in length and overlaps completely with ORF1 when read in the same direction. Site-directed mutagenesis was used to show that the ORF5 protein is not essential for PCV2 replication. To investigate the biological functions of the novel protein, we constructed a recombinant eukaryotic expression plasmid capable of expressing PCV2 ORF5. The results show that the GFP-tagged PCV2 ORF5 protein localizes to the endoplasmic reticulum (ER), is degraded via the proteasome, inhibits PAM growth and prolongs the S-phase of the cell cycle. Further studies show that the GFP-tagged PCV2 ORF5 protein induces ER stress and activates NF-kappa B, which was further confirmed by a significant upregulation in IL-6, IL-8 and COX-2 expression. In addition, five cellular proteins (GPNMB, CYP1A1, YWHAB, ZNF511 and SRSF3) were found to interact with ORF5 via yeast two-hybrid assay. These findings provide novel information on the identification and functional analysis of the PCV2 ORF5 protein and are likely to be of benefit in elucidating the molecular mechanisms of PCV2 pathogenicity. However, additional experiments are needed to validate the expression and function of the ORF5 protein during PCV2 infection in vitro before any definitive conclusion can be drawn.

Summary: Lineage infidelity, characterized by abnormal immature B lineage output, is rare but detectable in patients with MDS and non-CML MPN. Further investigation is needed to understand the underlying disease biology and its implications for patient management.

Antecedents and consequences of supply chain agility: a competence-capability-performance paradigm

Summary: This study investigates the impact of IT usage and employee multi-skilling on the agility of the supply chain. The findings suggest that internal agility is the foundation of supply chain agility and can improve supplier and customer agility. IT usage is more significant than employee multi-skilling in facilitating supply chain agility. Both internal and supplier agility positively affect supply chain performance, while customer agility has no significant impact.

Summary: Measurable residual disease (MRD) is a powerful prognostic factor in acute myeloid leukemia (AML). This study aims to identify pre-treatment molecular predictors of immunophenotypic MRD clearance in AML patients. The results showed that induction chemotherapy led to different MRD responses, with 35% achieving MRD- remission, 27% achieving MRD+ remission, and 38% having persistent disease. Subsequent therapy resulted in MRD conversion in 34% of MRD+ patients and 26% of patients with persistent disease. Specific gene mutations and karyotypic abnormalities were found to be associated with high or low rates of MRD- remission. Patients with fewer individual clones were more likely to achieve MRD- remission. Furthermore, the study demonstrated that achieving MRD- prior to allogeneic stem cell transplant (allo-SCT) was associated with favorable outcomes. Therefore, the inclusion of patients with specific baseline mutational patterns and high clone numbers in clinical trials should be considered.

Methodology and Implementation of Beam Steering Using C-Shaped Split Rings for Fabry-Perot Antennas

Summary: This article presents the design process of a beam-steering partially reflective surface (PRS) made up of C-shaped split-ring resonators (CSRRs). By analyzing the transmission characteristics of CSRRs with variable opening angles and orientations, spatially abrupt phase and magnitude are introduced over the PRS to reshape the wavefronts of linear polarization waves. Four arrangements of 5 x 5 CSRRs are fabricated to validate the design methodology of beam steering based on the designed two distributions and the two control methods. Furthermore, two reconfigurable Fabry-Perot (FP) antennas are proposed based on liquid-metal and mechanically rotating reconfiguration, demonstrating higher aperture efficiency, desirable radiation patterns, and excellent reconfiguration reliability. Moreover, an eight-element phased array combined with a liquid-metal PRS is fabricated and shows improved scanning performance and stable gain fluctuation.

Adenovirus infection diagnosed by metagenomic next-generation sequencing after haploidentical hematopoietic stem cell transplantation: A multicenter study in China

Summary: This study aims to observe and analyze the clinical characteristics and prognosis of adenovirus (ADV) infection diagnosed by metagenomic next-generation sequencing (mNGS) after haploidentical hematopoietic stem cell transplantation (Haplo-HSCT). The results showed that mNGS can provide screening for ADV and information on ADV subtypes, helpful for diagnosing patients at high risk for ADV infection. ADV infection can involve multiple organs, has difficulty in early diagnosis, and has a poor prognosis. Currently, effective treatments are inadequate.

In this study, we screened the anti-CSFV activity of 12 compounds targeting lipid metabolism pathways. 25-hydroxycholesterol (25HC) exhibited potent anti-CSFV activity by inhibiting virus entry into cells.

The effect of high-involvement human resource management practices on supply chain resilience and operational performance

Summary: This study explores the impact of high-involvement human resource management practices on supply chain resilience and the relationship between the dimensions of supply chain resilience and operational performance. The findings suggest that employee participation plays a crucial role in improving supplier, customer, and internal resilience. However, employee skills only facilitate internal and customer resilience, while employee incentives do not influence supply chain resilience. Additionally, internal and customer resilience have positive effects on operational performance, but supplier resilience does not significantly affect it. These findings contribute to both academia and practice.
