We report a kinetic study of the reaction between superoxide and an important intracellular form of vitamin B-12, cob(II)alamin. Superoxide is implicated in the pathophysiology of many inflammatory diseases, whereas vitamin B,2 derivatives are often beneficial in their treatment. We found that cob(II)alamin reacts with superoxide at rates approaching those of superoxide dismutase itself, suggesting a probable mechanism by which vitamin B-12. protects against chronic inflammation and modulates redox homeostasis.

The use of light sheet fluorescence microscopy (LSFM) allows for more precise measurements compared to traditional histological methods, providing a novel and robust 3D imaging platform for visualizing and quantifying arterial injury. LSFM outperforms classic histology in precision and quantitative capabilities while minimizing user bias associated with 2D histological analysis.

Summary: Staphylococcus aureus is a common human pathogen that often leads to chronic infections, with persister cells contributing to the failure of antibiotic treatment; host-derived peroxynitrite is identified as a major mediator of antibiotic tolerance in macrophages infected by S. aureus; these findings highlight the significant impact of host-bacterial interactions on antibiotic tolerance and suggest that modulating the host response could be a potential therapeutic strategy to enhance S. aureus sensitivity to antibiotics.

Antioxidant Response Activating nanoPartictes (ARAPas) localize to atherosclerotic plaque and locally activate the Nrf2 pathway

Summary: Atherosclerotic disease, driven by redox dysfunction, is a leading cause of death worldwide. However, the translation of redox-based therapies to clinical applications remains challenging. In this study, researchers developed Antioxidant Response Activating nanoparticles (ARAPas) that target the dysfunctional endothelium and selectively accumulate in atherosclerotic plaque. These nanoparticles encapsulate a redox-active drug and have anti-inflammatory and anti-oxidative effects. The results demonstrate the selective accumulation of these nanoparticles in atherosclerotic plaque and the activation of Nrf2-target genes.
