Summary: In this study, a facile and efficient method was developed to assemble dual-site-specific ADCs with defined DARs at both the N-glycosylation site and K248 site, either with the same payloads or with two types of payloads. The constructed dual-site ADCs showed acceptable homogeneity, excellent buffer stability, and enhanced in vitro and in vivo efficiency.

Summary: We present a traceless group directed one-pot regiospecific cycloaddition strategy for the synthesis of 4-fluorinated pyrroles, which have shown promising anti-inflammatory activity in previous studies. This method allows the synthesis of a variety of 4-fluoropyrroles that were previously inaccessible, and a proposed reaction mechanism is based on observed intermediates.

Summary: A one-pot four-component synthesis of diselenocarbamates was established under mild conditions, leading to the formation of 36 products in up to 91% yield. The zwitterionic intermediate formed from chloroform and amine was captured by diselane via nucleophilic substitution to afford the desired diselenocarbamate with the help of a base.

Summary: In this paper, a strategy for the facile and efficient one-step synthesis of site-specific antibody-drug conjugates (ADCs) using a thioester-based acyl transfer reagent is reported. This strategy allows for the selective modification of antibodies and demonstrates excellent homogeneity, stability, and anti-tumor activity.

Summary: We describe a method for the one-pot, three-component synthesis of sulfides using a sulfoxide reagent as a formal sulfur dication equivalent. This protocol involves three simple chemical operations with two Grignard reagents and trimethylsilyl chloride (TMSCl) to sequentially generate sulfenate anions, sulfenate esters, and sulfides. We demonstrate the versatility of this method by synthesizing a wide range of sulfides, including dialkenyl and alkenyl-alkynyl sulfides, without the need for thiols.

Summary: Heterotypic polyubiquitins, a newly emerging class of polyubiquitins, have attracted attention due to their diverse structures and physiological functions. However, the current methods for synthesizing heterotypic chains have limitations in terms of laborious processes and lack of modularity. In this study, a one-pot, photocontrolled synthesis method was established to construct structurally defined heterotypic polyubiquitin chains.

The biological production capability will enable low-cost, large-scale production and promote clinical applications of all-DNA nanomedicines.
