Vitiligo Skin T Cells Are Prone to Produce Type 1 and Type 2 Cytokines to Induce Melanocyte Dysfunction and Epidermal Inflammatory Response Through Jak Signaling

Christina Martins, Laure Migayron, Claire Drullion, Clement Jacquemin, Fabienne Lucchese, Jerome Rambert, Ribal Merhi, Pauline Michon, Alain Taieb, Hamid-Reza Rezvani, Emanuele de Rinaldis, Julien Seneschal, Katia Boniface

Summary: Vitiligo is a complex skin disorder involving both immune responses and melanocyte dysfunction. In addition to being targeted by T cells, melanocytes may actively contribute to the inflammation process. Jak inhibitors have the potential to prevent the impact of T cells on skin cells and pigmentation, offering clinical benefits for vitiligo treatment.

The humanistic burden of vitiligo: a systematic literature review of quality-of-life outcomes

M. Picardo, R. H. Huggins, H. Jones, R. Marino, M. Ogunsola, J. Seneschal

Summary: This systematic literature review provides comprehensive evidence for the humanistic burden in patients with vitiligo. The study finds that vitiligo has a significant negative impact on the quality of life of patients and their families, and treatment can significantly improve patient's quality of life. Factors such as female sex, visible lesions, and lesions in sensitive areas are associated with reduced quality of life.

An integrated analysis of herpes virus infections from eight randomized clinical studies of baricitinib in adults with moderate-to-severe atopic dermatitis

T. Werfel, A. D. Irvine, C. Bangert, J. Seneschal, S. Grond, T. Cardillo, D. Brinker, J. Zhong, E. Riedl, A. Wollenberg

Summary: This study examined the occurrence of herpes simplex infections in adult patients with AD receiving baricitinib treatment. The study found that herpes simplex was common, eczema herpeticum occurred less frequently, and herpes zoster events were rare and not dose-dependent. Most herpes simplex events were mild or moderate, and rarely led to permanent discontinuation.

Reynolds, Carsten Flohr, Phyllis Spuls

Summary: The TREAT Registry Taskforce is a collaborative network collecting real-world data on atopic eczema patients. This study provides an overview of the eight established TREAT registries and examines the overlap and pooling ability of their datasets. The results show that pooled analyses can be performed on multiple important domain items, enabling future comparative analyses.

Impact of house dust mite in vitiligo skin: environmental contribution to increased cutaneous immunity and melanocyte detachment

Hanene Bzioueche, Katia Boniface, Claire Drullion, Sandrine Marchetti, Berengere Chignon-Sicard, Laura Sormani, Stephane Rocchi, Julien Seneschal, Thierry Passeron, Meri K. Tulic

Summary: Vitiligo is an autoimmune disease characterized by melanocyte loss and white patches on the skin. This study demonstrates that house dust mite (HDM) can induce inflammatory responses and melanocyte detachment in vitiligo through protease activity. HDM may act as an external source of pathogen-associated molecular pattern molecules in vitiligo, and MMP-9 inhibitors could be potential therapeutic targets. Furthermore, studying specific vitiligo endophenotypes can help understand the environmental contribution to the disease.

Vitiligo patient population and disease burden in France: VIOLIN study results from the CONSTANCES cohort

Khaled Ezzedine, Julien Seneschal, Audrey Da Silva, Nathalie Preaubert, Anne Lamblin, Carine Delattre, Corinne Emery, Camille Nevoret, Jonathan Finzi, Stephane Bouee, Thierry Passeron

Summary: This study assessed the prevalence, disease burden, and treatment of vitiligo in France. The results showed that the prevalence of vitiligo in France was 0.71%, most patients were diagnosed before the age of 30, and had visible skin lesions. Vitiligo patients had a high burden of comorbidities and impaired quality of life, but the majority of patients did not receive any prescribed treatment.
