Summary: In this study, a LC-MS/MS method for the quantitation of fructose in human urine was developed and validated. Difficulties were encountered for the post-extraction stability experiments, particularly for authentic fructose QCs at low concentrations. However, a column cleaning procedure was implemented to overcome this issue. The validated HILIC-MS/MS method was successfully applied to analyze clinical samples with a 91% overall run passing rate.

Summary: Oligonucleotides (OGNs) are a relatively new modality that provides unique opportunities for expanding therapeutic targets. Liquid chromatography-mass spectrometry (LC-MS) is now becoming the preferred method for bioanalysis of OGNs. While ion pairing reversed-phase liquid chromatography (IP-RPLC) has been widely used, there are technical issues associated with these methods. IP-free methods, such as hydrophilic interaction liquid chromatography (HILIC) and anion-exchange techniques, have emerged as promising approaches. In this review, the state-of-the-art IP-RPLC-MS bioanalytical methods for OGNs and their metabolites published in the past 10 years (2012-2022) are critically reviewed. Recent advances in IP-reagent-free LC-MS bioanalysis methods are discussed. Finally, future opportunities for developing new methods for comprehensive bioanalysis of OGNs are described.

Summary: This study proposed and validated a novel strategy of one sample multi-point calibration curve (OSCC) using multiple isotopologue reaction monitoring (MIRM) for quantitative LC-MS bioanalysis. The developed OSCC methodology exhibited good performance in selectivity, sensitivity, accuracy, and precision, and could realize sample dilution by monitoring the MIRM channel. The advantages of excluding the preparation of multi-sample external calibration curve (MSCC) and sample dilution in OSCC make this strategy widely applicable in various fields.

Summary: Therapeutic drug monitoring (TDM) of antibiotics is crucial in high pharmacokinetic variability populations like critically ill patients. A new method for simultaneous quantification of ten antibiotics using 5-sulfosalicylic acid dihydrate (SSA) solution and 2D-LC-MS/MS was developed and evaluated in a one-year retrospective study. The method showed good assay sensitivity, eliminated >90% of plasma proteins, and allowed rapid dosage adjustment.

Summary: An uncommon enzymatic-fingerprinting workflow was proposed for the characterization and discrimination of mushroom polysaccharides using HILIC-ESI-MS. The study revealed the structural features of polysaccharides in Hericium erinaceus fruiting bodies and visually classified different polysaccharides through similarity analysis and other methods. This enzymatic-fingerprinting workflow provides an effective example for quality evaluation of fungi polysaccharides using a structural-fingerprinting strategy.

Summary: This study aimed to evaluate different combinations of polyanion-metal ion based solution system for phospholipids removal and elimination of matrix effects in human plasma. The optimized ZrOCl2-Ludox/DSS systems provided efficient phospholipids removal and adequate analyte recovery, and they also correctly eliminated ion suppression or enhancement of the non-polar and polar drugs.
