Additionally, the study found that NGS of CSF ctDNA may provide more accurate information about intracranial lesions compared to ctDNA in blood serum, indicating a potentially better detection method.

These findings support further exploration of combined BRAF inhibitors and actin-targeting drugs as a potential therapeutic approach in BRAFV600E-mutated cancers.

Vitamin D Modulates the Response of Patient-Derived Metastatic Melanoma Cells to Anticancer Drugs

Summary: Preconditioning of patient-derived melanoma cells with 1,25(OH)(2)D-3 enhances the inhibitory effect of cediranib and vemurafenib and reduces mitochondrial respiration parameters. Moreover, cancer-associated fibroblasts play a crucial role in melanoma growth. Thus, 1,25(OH)(2)D-3 could be considered as an adjuvant agent in the treatment of malignant melanoma.

Strategic substitution of fluorine at the C5 position increases solubility and enables oral dosing, leading to significant tumor growth inhibition in combination with the MEK inhibitor cobimetinib in a KRAS mutant xenograft mouse model.

Summary: In mouse models of BRAF- and NRAS-mutant melanoma, upfront immunotherapy enhances responses to targeted therapy. The sequence of IT -> TT modulates the immune environment, increasing infiltration of T cells and decreasing tumor-associated macrophages, leading to improved therapeutic outcomes. Durable responses to the IT -> TT sequence depend on T-cell activity and enrichment of a population of melanoma cells with increased expression of MHC class I and melanoma antigens.

Discovery of a highly potent pan-RAF inhibitor IHMT-RAF-128 for cancer treatment

Summary: In this study, a highly potent pan-RAF inhibitor, IHMT-RAF-128, was developed, which demonstrated strong anti-tumor efficacy against cancer cells harboring RAF or RAS mutations, especially the KRAS-G12C secondary mutations resistant to AMG510. IHMT-RAF-128 also exhibited excellent pharmacokinetic profile and dose-dependent anti-tumor efficacy in xenograft mouse tumor models without obvious toxicities. These findings support further investigation of IHMT-RAF-128 as a potential clinical drug candidate for cancer patients with RAF or RAS mutations.

Summary: This study explores the relationship between MGMT promoter methylation and the sensitivity of glioblastoma patients to temozolomide chemotherapy, finding that a higher percentage of MGMT promoter methylation is associated with smaller tumor volume, lower fluorescence rate with 5-ALA, and greater extent of resection. Higher MGMT promoter methylation is also related to improved progression-free survival and overall survival, even after adjusting for extent of resection. Additionally, a higher number of chemotherapy cycles is associated with longer progression-free survival and overall survival.

The importance of histological diagnosis prior to treatment of sellar or suprasellar lesions with atypical or aggressive features is highlighted. The patient underwent surgery and palliative whole brain radiotherapy, but died four months later.
