Summary: In this study, liver-specific Pcx knockout mice (PcxL-/-) were generated to investigate the role of Pcx in hepatic mitochondrial metabolism under different physiological states. The results showed that hepatic Pcx is important for maintaining long-term glycemia under carbohydrate-limited diets, and loss of Pcx is associated with changes in the abundance of acetylated proteins.

Summary: Recent studies have shown that renal gluconeogenesis is stimulated in the presence of obesity, and this stimulation is potentially regulated by Atp6ap2 via the PGC1-alpha/AKT-1 pathway in response to high fat diet. The upregulation of renal gluconeogenesis leads to increased expression of PEPCK and G6Pase, while Atp6ap2-KO can reverse these changes.

This study investigates novel insulin analogues with different chemical compositions to understand signaling bias and finds that analogues with an A6-A11 cis-dicarba bond have low mitogenic potency. These findings will assist in developing insulin analogues for diabetes treatment with low mitogenic activity and reduced risk of promoting cancer.

Summary: Flavin adenine dinucleotide (FAD) interacts with flavoproteins to mediate oxidation-reduction reactions for cellular energy demands. Depleting FAD pools in mice causes phenotypes associated with organic acidemias and other inborn errors of metabolism (IEMs), such as reduced body weight, hypoglycemia, and fatty liver disease. The study reveals that B2D inhibits fasting activation of PPAR alpha target genes and downregulates gluconeogenesis genes. Knockdown of PPAR alpha in the liver recapitulates B2D effects, while treatment with the PPAR alpha agonist fenofibrate rescues fasting glucose availability and overcomes B2D phenotypes.
