In this work, pollen mother cells (PMCs) isolated in Arabidopsis thaliana by laser-capture microdissection (LCM) were subjected to transcription profiling by microarray (LMM). PMCs covering all meiotic stages, from leptotene to tetrad, were collected in an Atmcc1 characterized by overexpression of a GCN5-like histone acetylase (AtMCC1). A total of 150 genes showed differential expression in Atmcc1 PMCs as compared to the wild type. Histone hyperacetylation affected the transcription of genes belonging to categories such as the meiotic and mitotic cycle, the ubiquitinproteasome-system, and the chromatin structure. We also discuss the putative role of ASK1 and RAD51C upregulation in the meiotic defects observed in Atmcc1. PMCLMM experiments allowed identification of candidate AtMCC1 targets with known and potential function in meiosis, providing data for further investigation on plant meiosis.

Giuseppina Bruno, Valeria Li Bergolis, Annamaria Piscazzi, Fabiana Crispo, Valentina Condelli, Pietro Zoppoli, Francesca Maddalena, Michele Pietrafesa, Guido Giordano, Danilo Swann Matassa, Franca Esposito, Matteo Landriscina

Summary: The study reveals the role of the TRAP1 gene in the adaptive response of human colorectal cancer cells to hypoxia. TRAP1 is involved in regulating HIF-1α stabilization, glycolytic metabolism, and gene expression reprogramming. It also affects glucose uptake, lactate production, and ribosome biogenesis through the mTOR pathway.

Summary: Understanding the recombination behavior in polyploidy species is crucial for genetic discoveries. This study focused on blueberry, a tetraploid species, and investigated its genetic behavior using cytogenetics and high-density genetic maps. The study revealed the presence of an autotetraploid behavior and a translocation affecting chromosome pairing and recombination. No other structural genomic divergences were detected, providing valuable resources for future genetic and comparative genomic studies.

Summary: This study aimed to evaluate the cross-talk among metabolism, stress response, immune system and reproduction in immature and mature female swordfish. Transcriptomic and histological analysis revealed that mature females have compromised detoxification capacity and immune system, indicating higher susceptibility to environmental stress and pollutants during the reproductive season.

Summary: The occurrence of Staphylococcus microti (S. microti) in milk and milking parlor equipment samples of an Italian buffalo farm was assessed in this study. The results showed that S. microti was commonly found in buffalo milk and on milking parlor surfaces, with 20.4% of the isolates exhibiting multidrug resistance. This study highlights the importance of milking parlor environmental quality in the spread of S. microti and the microbial communities in milk.

Comparison of Tomato Transcriptomic Profiles Reveals Overlapping Patterns in Abiotic and Biotic Stress Responses

Ciro Gianmaria Amoroso, Daniela D'Esposito, Riccardo Aiese Cigliano, Maria Raffaella Ercolano

Summary: This study analyzed and compared the transcriptomic responses of resistant and susceptible tomato genotypes to multiple biotic and abiotic stresses. Many genes involved in defense mechanisms were identified, including those encoding for TFs, phytohormones, and participating in signaling and cell wall metabolic processes. A total of 1474 DEGs were commonly found between biotic and abiotic stress, with 67 DEGs involved in response to at least four different stresses. The detected genes responsive to multiple stress could be further studied and utilized to enhance plant tolerance through biotechnological approaches.

Summary: This study investigated the potential impact of a co-feed liquid whey-integrated diet on the fecal microbiota of crossbred pigs. The results showed that the supplementation with liquid whey affected the diversity and abundance of the bacterial community in the pig's feces. Furthermore, the correlation analysis suggested that the identified differentially abundant genera were associated with the physiological parameters of the pigs.

Programmed Cell Death in Sea Urchins: A Review

Viviana Di Tuccio, Pasquale De Luca, Giovanna Romano

Summary: The sea urchin embryo is a widely used model system to study biological processes and investigate molecular and cellular responses to external stress factors. Programmed cell death (PCD) is a regulated molecular mechanism that plays a central role in tissue rearrangement and development, especially during metamorphosis, and is activated in response to abiotic stress. This review summarizes the current knowledge about the involvement of PCD processes in sea urchin embryos exposed to pollutants and provides information about the occurrence of physiological apoptosis during development.

A novel telomerase activity and microRNA-21 upregulation identified in a family with palmoplantar keratoderma

Bruna De Felice, Concetta Montanino, Claudia Pinelli, Massimo Nacca, Pasquale De Luca

Summary: Palmoplantar keratoderma is a group of skin diseases characterized by thickening of the palms and soles. Various genetic mutations, such as KRT9, KRT1, AQP5 and SERPINB 7, have been identified as triggers for palmoplantar keratoderma. In this study, a family with palmoplantar keratoderma caused by autosomal dominant KRT1 mutations was analyzed for KRT1 genetic sequence, telomerase activity, and miR-21 expression. The patients showed thickening of the skin on their hands and feet, KRT1 mutations, and high expression levels of hTERT, hTR, and miR-21, indicating aberrant proliferation and inflammation.

Novel Insights on the Bacterial and Archaeal Diversity of the Panarea Shallow-Water Hydrothermal Vent Field

Erika Arcadi, Emanuela Buschi, Eugenio Rastelli, Michael Tangherlini, Pasquale De Luca, Valentina Esposito, Rosario Calogero, Franco Andaloro, Teresa Romeo, Roberto Danovaro

Summary: Current knowledge of the microbial diversity of shallow-water hydrothermal vents is limited. This study used 16S rRNA gene metabarcoding to investigate the bacteria and archaea in seawater and sediments of three shallow-water hydrothermal systems. The results showed significant differences in microbial diversity between seawater and sediments. Additionally, the benthic microbial assemblages differed markedly among the three areas, reflecting the different hydrothermal activities. Overall, this study suggests that hydrothermal vent dynamics contribute to an increase in microbial diversity in this unique system.

Summary: The methylation profile of HSPCs following AHSCT was analyzed and found to undergo changes that may be useful in transplant diagnostics. The methylation signature was also found to be related to patient outcome. This study suggests that the methylation profile of HSPCs can provide prognostic information for engraftment success and graft failure prediction in AHSCT.
