Gastroprotective activity of ferruginol in mice and rats:: effects on gastric secretion, endogenous prostaglandins and non-protein sulfhydryls

The gastroprotective mechanism of the natural diterpene ferruginol was assessed in mice and rats. The involvement of gastric prostaglandins (PGE(2)), reduced glutathione, nitric oxide or capsaicin receptors was evaluated in mice either treated or untreated with indometacin, N-ethylmaleimide (NEM), N-nitro-L-arginine methyl ester (L-NAME) or ruthenium red, respectively, and then orally treated with ferruginol or vehicle. Gastric lesions were induced by oral administration of ethanol. The effects of ferruginol on the parameters of gastric secretion were assessed in pylorus-ligated rats. Gastric PGE(2) content was determined in rats treated with ferruginol and/or indometacin. The reduction of gastric glutathione (GSH) content was determined in rats treated with ethanol after oral administration of ferruginol, lansoprazole or vehicle. Finally, the acute oral toxicity was assessed in mice. Indometacin reversed the gastroprotective effect of ferruginol (25 mg kg(-1)) but not NEM, ruthenium red or L-NAME. The diterpene (25 mg kg(-1)) increased the gastric juice volume and its pH value, and reduced the titrable acidity but was devoid of effect on the gastric mucus content. Ferruginol (25, 50 mg kg(-1)) increased gastric PGE(2) content in a dose-dependent manner and prevented the reduction in GSH observed due to ethanol-induced gastric lesions in rats. Single oral doses up to 3 g kg(-1) ferruginol did not elicit mortality or acute toxic effects in mice. Our results showed that ferruginol acted as a gastroprotective agent stimulating the gastric PGE(2) synthesis, reducing the gastric acid output and improving the antioxidant capacity of the gastric mucosa by maintaining the GSH levels.

Summary: This study reports a green extraction method for lichen substances from Hypotrachyna cirrhata using ethyl lactate and high power ultrasounds. Phenolic Composition and alpha-Glucosidase Inhibition of Leaves from Chilean Bean Landraces

Summary: In this study, the MeOH:H2O (7:3) extracts of leaves from Chilean bean landraces were evaluated for their antioxidant activity and inhibition of enzymes associated with metabolic syndrome. The results showed that these extracts exhibited high antioxidant activity and strong inhibition against alpha-glucosidase. Caffeoylmalic acid and flavonol glycosides were identified as the main active compounds. The leaf extracts of Chilean beans have the potential to be a source of bioactive compounds.

Summary: In this study, the chemical composition and biological activity of Ovidia pillopillo, an endemic species of the Valdivian Forest in Chile, were investigated for the first time. The plant was found to contain various flavonoids and coumarins, and exhibited strong antioxidant and enzyme inhibition activities. These findings suggest that Ovidia pillopillo has potential as a source of new medicines.

Summary: In this study, hyphenated liquid chromatography with high-resolution mass spectrometry was used to analyze the metabolomes of eleven species of Sticta lichens. Chemical markers for fast identification of Sticta lichens were identified for the first time. The study also compared chemical compounds and traditional morphological traits in the study of ancestor-descendant relationships in the genus, and found inconsistencies between the two methods.

Summary: Lichens are symbiotic organisms that produce secondary compounds. Using hyphenated techniques, 25 compounds were detected from Antarctic lichen, including two new compounds and 10 phenolic compounds that were found for the first time in the Cladonia genus.

Summary: This study analyzed a traditional recipe used by the Mbya culture in Paraguay to treat cancer with the native toad Rhinella schneideri. The composition of the recipe was determined and its effects on human breast cancer cells were evaluated. The results showed that the recipe contains various metabolites, including alkaloids and bufadienolide derivatives. The recipe did not have cytotoxic effects on breast cancer cells, but it increased mitochondrial bioenergetics and inhibited cell migration.

Summary: In this study, the metabolite fingerprinting, antioxidant, and enzyme inhibitory potential of Himantormia lugubris, a small lichen shrub native to Chile and growing in the Antarctica region, were investigated for the first time. The results showed that this species is a rich source of bioactive compounds with good inhibition activity against cholinesterase and tyrosinase enzymes, as well as potential antioxidant activity.

A sustainable application for the extraction of lichen metabolites from Usnea cornuta: nontargeted metabolomics and antioxidant activity

Summary: This study isolated and purified lichen substances from Usnea cornuta using conventional solvents, green solvents, and green technologies. By using UHPLC/ESI/MS/MS, several lichen compounds were identified, and the quantification of usnic acid, diffractaic acid, and galbinic acid was performed. The extraction properties and antioxidant capacities of limonene, ethyl lactate, and methanol were compared, and the results showed that ethyl lactate could replace methanol as a more sustainable green chemistry method with similar antioxidant capacity.

Chemical profile and bioactivity of Chilean bean landraces (Phaseolus vulgaris L.)

Summary: This study aimed to analyze the chemical composition, antioxidant capacity, and enzyme inhibition effect of secondary metabolites-enriched extracts (SMEE) of thirteen Chilean bean landraces before and after boiling. HPLC-DAD-MS/MS was used to determine the SMEE composition, and various compounds including procyanidins, flavonoids, phenylpropanoids, and saponins were identified. Cooking decreased the total phenolics and saponins content, but certain individual compounds like ferulic acid and soyasaponins Ba and Bb increased. Total phenolics showed a significant correlation with antioxidant activity, and some saponins exhibited a correlation with α-glucosidase inhibition. The findings suggest that bean components may have local activity in the intestine, preventing oxidation of other nutrients and modulating gastrointestinal enzymes.

Antioxidant capacity and inhibition of metabolic syndrome-associated enzymes by Cryptocarya alba fruits

Summary: This study investigated the phenolic content, antioxidant capacity, and inhibitory effects on enzymes related to metabolic syndrome of the peumo fruits consumed by Mapuche Amerindians in Chile. The results showed that peumo fruits contained beneficial compounds and had significant effects on enzyme activity and antioxidant capacity. Furthermore, significant differences were observed in the fruit parts and processing methods.

Changes in polyphenol composition, antioxidant capacity and enzyme inhibition in Phaseolus vulgaris L. submitted to hydric stress

Summary: The aim of this study was to assess the impact of hydric stress on the phenolic composition, antioxidant capacity, and inhibition of enzymes associated with metabolic syndrome, in nine bean landraces cultivated in the Mediterranean zone of Chile. Results showed that under restricted watering, all phenolic compounds increased in Pinto beans and decreased in other accessions. Best effect towards alpha-glucosidase was observed for Negro Argel, Bayote, and Enriqueta with IC50 between 0.006 μg/mL and 0.820 μg/mL. Water stress has a significant effect on polyphenols composition and consequently on the antioxidant activity and inhibition of enzymes.

The importance of active surveillance of carbapenem-resistant Enterobacterales (CRE) in colonization rates in critically ill patients

Summary: This study emphasizes the importance of active surveillance for carbapenem-resistant Enterobacterales (CRE) and evaluates the prevalence of invasive infections, risk factors, and mortality risk in CRE-colonized patients. The results reveal that early detection of CRE colonization through screening testing is a vital tool for controlling CRE spread, but there is ineffective control of colonization and infection. The study highlights the significance of implementing preventive measures and improving teamwork.
