In Situ Gel Formulation for Enhanced Ocular Delivery of Nepafenac

Nepafenac is a water-insoluble nonsteroidal antiinflammatory drug that is available as an ophthalmic suspension (Nevanac (R)). Suspensions are undesirable for 2 reasons: they tend to cause foreign body sensation and lacrimation, which could limit residence time and drug bioavailability. This decreases the amount of time the drug has to reach the site of action, the cornea. Previously, we improved the solubility and ocular permeability of nepafenac by complexing the drug with hydroxypropyl-beta-cyclodextrin. In this study, we used the complex to formulate an ion-activated in situ gel system using sodium alginate, Protanal PH 1033, to increase the residence time and to reduce repeat eye drop instillation. Rheological properties of the formulations revealed that the viscosity of the optimized formulation was increased 30-fold when exposed to the simulated tear fluid (35 degrees C). Permeation studies showed that the drug concentration of the in situ formulations were approximately 10 times higher than the commercial product, Nevanac (R) (p < 0.001). In addition, the in situ gel formulations had 5-fold higher concentrations of nepafenac retained in the cornea when compared to Nevanac (R) (p < 0.001). Finally, ex vivo drug distribution studies in the porcine eye perfusion model revealed a higher drug retention in various ocular tissues such as cornea, sclera, retina, as compared to Nevanac (R). Cardiolipin for Enhanced Cellular Uptake and Cytotoxicity of Thermosensitive Liposome-Encapsulated Daunorubicin toward Breast Cancer Cell Lines

Summary: This study developed a co-delivery system of daunorubicin and cardiolipin encapsulated in thermosensitive liposomes for the treatment of breast cancer. The liposomes exhibited high drug encapsulation efficiency, small size, and rapid release under mild hyperthermia, resulting in significantly higher cytotoxicity against breast cancer cells compared to daunorubicin or conventional liposomes. This research lays the foundation for future development of liposomal co-delivery systems for breast cancer therapy.

Summary: This study investigates the preparation of an inclusion complex of Hispolon with Sulfobutylether-beta-cyclodextrin (SBE beta CD) and the formulation of Hispolon-SBE beta CD trapped within sterically stabilized liposomes (HSC-SL). The results show that HSC-SL exhibits enhanced anticancer activity against melanoma cell lines and sustained drug release compared to Hispolon liposomes and Hispolon-SBE beta CD liposomes.

Summary: Microwave-assisted synthetic methods are commonly used for the surface modification and functionalization of multi-walled carbon nanotubes (MWCNTs) in diverse drug delivery applications. Microwave-induced functionalization of MWCNTs offers high efficiency and shorter reaction time compared to conventional techniques. Microwave methods are simple, fast, and effective for the covalent and noncovalent conjugation of MWCNTs with various biomolecules and polymers. This review focuses on the synthesis and drug delivery applications of microwave irradiation techniques (MITs) for the functionalization of MWCNTs using amino acids, vitamins, proteins, epoxy moieties, metal nanoparticles, and polymers.

Topical Delivery of Diacetyl Boldine in a Microemulsion Formulation for Chemoprotection against Melanoma

Summary: This study aimed to develop a microemulsion formulation for topical delivery of Diacetyl Boldine (DAB) and evaluate its cytotoxicity against melanoma cell line (B16BL6) in vitro. The results showed that the microemulsion formulation exhibited significantly higher skin retention levels than the DAB solution in MCT oil. Additionally, the microemulsion formulations showed substantially higher cytotoxicity toward B16BL6 cell lines than the control formulation.

Liposomal DQ in Combination with Copper Inhibits ARID1A Mutant Ovarian Cancer Growth

Summary: Therapeutic strategies for ARID1A-mutant ovarian cancers are limited. Higher basal ROS and lower basal GSH enhance the aggressive proliferation ability and metastatic property of OCCCs through EMT induction and immunosuppressive microenvironment. However, the aberrant redox homeostasis also increases the sensitivity of DQ-Lipo/Cu in mutant cells. DQ generates DDC in response to ROS, which together with Cu generates ROS cascade and targets GSH vulnerability, disrupting redox homeostasis and causing cancer cell death. The formed Cu(DDC)2 acts as a potent cytotoxic anti-cancer drug inducing immunogenic cell death, providing a promising strategy for managing cancer metastasis and drug resistance.

Comparison of the use of a standard cytology brush versus a mini cytology brush to obtain conjunctival samples for cytologic examination in healthy dogs

Summary: The study compared the quality of conjunctival samples obtained with two different brushes for cytologic examination in healthy dogs. Results showed that samples collected with a standard cytology brush had better cellularity, less background debris, and more uniform distribution of cells, while samples collected with a mini cytology brush had less cellular overlapping and mucus in the background.
