Summary: Stable isotope tracers are commonly used with gas chromatography coupled to mass spectrometry for enzyme activity and metabolic pathway studies. In this research, three commercially available instruments were used to analyze samples from a stable isotope labeling study on Rothia mucilaginosa metabolism. The study found that all three GC-MS instruments can be used for stable isotope tracing studies, with high-resolution GC-QTOF MS offering additional capabilities in identifying the chemical structures of unknown compounds.

Summary: An effective LC x LC/Q-TOF MS method was developed for quantitative determination of 19 active constituents in chrysanthemum species. Principal component analysis, partial least squares discriminant analysis, and hierarchical cluster analysis were used to distinguish the chrysanthemum varieties. LC x LC/Q-TOF MS combined with PCA, PLS-DA, and HCA can be successfully applied for the classification and quality control of chrysanthemum.

Summary: A workflow for comprehensive semi-polar metabolite profiling was developed and validated using liquid chromatography coupled with electrospray ionization ion mobility quadrupole time-of-flight mass spectrometry. This workflow has the capability to distinguish different cultivars of Hydrangea macrophylla ssp. serrata and obtain a list of metabolites responsible for the differentiation. Suspect screening of phenyldihydroisocoumarins provides valuable information for the biosynthesis of phyllodulcin.

Summary: In this study, the metabolism of celastrol in vivo and in vitro was comprehensively characterized using ultra-high-performance liquid chromatography coupled with hybrid triple quadrupole time-of-flight mass spectrometry. A total of 43 metabolites were identified and characterized, and it was found that the biotransformation capacity of the intestinal tract exceeded that of the liver. These findings provide a basis for understanding the pharmacological mechanisms of celastrol and have potential implications for clinical applications.

Summary: A rapid and sensitive LC-QTOF method was developed for determining resin acid concentrations in aqueous pulp and paper effluent samples. The method demonstrated excellent calibration R-2 values for twelve resin acids and achieved a low limit of quantitation without complex sample extraction. High precision and accuracy results were obtained for specific resin acids, with detection limits ranging from 0.05 to 0.07 μg/L. The method showed clear advantages over traditional gas chromatography and capillary electrophoresis methods.

Summary: A unique and effective two-dimensional liquid chromatography system was established for the analysis of bioactive components in honeysuckle. The system utilized optimized conditions, columns, flow rates, and organic solvents to improve chromatographic resolution. 57 compounds were identified by mass spectrometry, and it was found that the composition of honeysuckle varied significantly among different regions. Additionally, most samples exhibited potent alpha-glucosidase inhibitory activity, which is beneficial for drug quality evaluation.

Summary: This research focused on the biodegradation of synthetic estrogenic compound 17 alpha-ethinylestradiol (EE2) and compared its degradation efficiency and power output in suspension flasks and a Bioelectrochemical System (BES). The results showed that the BES had significantly higher EE2 removal rate and power density. The study also identified a potential pathway for EE2 degradation using the newly isolated strain Rhodopseudomonas palustris MDOC01, and detected important metabolites during the bioelectrochemical treatment process of EE2.

Summary: This study characterized the volatile aroma of bergamot using headspace solid-phase microextraction coupled with comprehensive two-dimensional chromatography quadrupole time-of-flight mass spectrometry. Gas chromatography-olfactometry was used to characterize the aroma-active compounds. A total of 323 volatile compounds and 29 aroma-active compounds were identified. Comprehensive two-dimensional gas chromatography quadrupole time-of-flight mass spectrometry detected 218 more compounds than gas chromatography-mass spectrometry due to higher peak capacity and separation resolution. The key characteristic volatiles of bergamot included alpha-terpineol, isobornyl formate, alpha-phellandrene dimer, furfural, and o-acetyltoluene. The volatile profiles can be used as a reference for the identification and quality appraisal of bergamot and its products.

The study found higher alkaloid concentrations in the inner section of stems compared to the outer surface, with side branches showing significantly higher alkaloid content than the main stem, indicating a gradient decrease of alkaloids towards branch tips.

Summary: In this study, the quality of Yiqing tablet was comprehensively evaluated using multi-component quantification, multi-dimensional fingerprint construction, and antioxidant activity analysis. The results showed that the sample had 25 components with antioxidant activity according to the HPLC fingerprint and most bands of FTIR also showed antioxidant activity. Based on the evaluation results of HPLC and FTIR fingerprint, all samples were classified as first level, demonstrating the consistency of sample quality. This study provides a new idea for achieving comprehensive quality evaluation of traditional Chinese medicines.

The optimized experimental parameters result in low dissolved organic carbon content and residual acidity concentration. The accuracy of the method is confirmed through the analysis of certified reference materials, and its environmental friendliness is validated using the AGREE metric. Principal component analysis is used to classify the medicinal herbs based on their inorganic constituent concentrations.

Summary: This study investigated the cellular uptake of low, medium and high molecular weight polyethylene glycol (PEG) in MCF-7 cells using UPLC-MS/MS and CID techniques. The results showed that PEG600, PEG2K, PEG4K and PEG10K exhibited low toxicity and had limited cellular entry at concentrations of 5-1200 μg/mL. Additionally, the environmental friendliness of the developed analytical methods was evaluated.

The results highlighted the potential of steroidal saponins, isoflavones, and flavones as active constituents of fenugreek with implications for lactation, reproductive processes, and estrogenic activity. Animal model study further supported the presence of these bioactive compounds and their effects on body weight, serum prolactin levels, serum estradiol levels, and ovarian histological structure.
