Chromatography-tandem mass spectrometry method for the simultaneous quantitation of metoprolol succinate and simvastatin in human plasma

The liquid chromatographic-tandem mass spectrometry method was developed for the accurate quantitation of metoprolol succinate (MET) and simvastatin (SIM) in human plasma which were obtained from the pharmacokinetic (PK) study. The sample purification and pre-concentration was performed by protein precipitation technique using propranolol hydrochloride as working internal standard (WIS). The chromatographic separation was achieved using an isocratic mobile phase consisting of a mixture of acetonitrile and 0.5% formic acid (90: 10 (v/v), pH 3.5) flowing through C18 column at a flow rate of 0.2 ml/min. Electro spray ionization (ESI) with multiple reaction monitoring (MRM) was used to acquire mass spectra. Ions were monitored in positive mode and the mass transitions measured were m/z 268.1 -> m/z 103.2, m/z 441.3 -> m/z 325.1 and m/z 260.0 -> m/z 129.5 for MET, SIM and WIS, respectively. An extensive pre-study method validation was carried out in accordance with USFDA guidelines. The linearity for the calibration curve in the concentration range of 1.0-500.0 and 0.1-20 ng/ml for MET and SIM, respectively and the lower limits of quantitations (LLOQ) were 1.0 and 0.1 ng/ml for MET and SIM, respectively. The method was successfully applied to a PK study on fixed dose combination (FDC) tablet containing MET and SIM in healthy human subjects. (C) 2009 Elsevier B.V. All rights reserved.

Summary: Rheumatoid arthritis (RA) is a severe inflammatory disease that causes joint swelling, stiffness and pain, posing a significant global threat. Damage-associated molecular patterns (DAMPs) are endogenous danger molecules that are released during cell injury or death, activating various inflammatory diseases. EDA-fibronectin (Fn) is a DAMP molecule responsible for triggering RA through its interaction with TLR4. This study aimed to reveal the interaction of other PRRs with EDA-Fn in RA through computational methods and identified TLR2, TLR5, and RAGE as potential PRRs.

Summary: The study found that Tragia involucrata has potential anti-inflammatory effects and can reduce symptoms of arthritis and restore normal tissue structure. Further analysis revealed the presence of flavonoids such as gallic acid, rutin, and quercetin in the plant, which showed potent anti-inflammatory potential. These findings suggest that Tragia involucrata could be a potential therapeutic candidate for treating inflammatory diseases.

Summary: The anti-osteoporotic effect of Methanolic extract of Oldenlandia umbellata (MOU) was evaluated in MG-63 cells and RANKL-stimulated RAW 264.7 cells. MOU promoted osteoblast differentiation and inhibited osteoclast formation through regulating multiple signaling pathways. These findings highlight the potential of MOU as a therapeutic agent for the treatment of osteoporosis.

In Silico Rationalization for Leads from Oldenlandia umbellata L. to Inhibit Multiple Molecular Targets Regulating Osteoporosis

Summary: This study computationally screened the phytochemical constituents of Oldenlandia umbellata Linn against the therapeutic targets of osteoporosis and validated the results using molecular dynamics simulation. The results showed that deacetylasperuloside, a compound found in Oldenlandia umbellata Linn, exhibited good drug-likeness and bioactive scores and could potentially bind with the molecular targets of osteoporosis. Molecular dynamic simulation further confirmed the stability of the ligand-protein complexes involving deacetylasperuloside.

A strategy takes Yiqing tablets as an example to carry out simpler multi-component quantification and use fingerprint technology for comprehensive quality consistency evaluation

Summary: In this study, the quality of Yiqing tablet was comprehensively evaluated using multi-component quantification, multi-dimensional fingerprint construction, and antioxidant activity analysis. The results showed that the sample had 25 components with antioxidant activity according to the HPLC fingerprint and most bands of FTIR also showed antioxidant activity. Based on the evaluation results of HPLC and FTIR fingerprint, all samples were classified as first level, demonstrating the consistency of sample quality. This study provides a new idea for achieving comprehensive quality evaluation of traditional Chinese medicines.

Green sample preparation of medicinal herbs in closed digester block for elemental determination by ICP OES

Summary: This work presents a simple, cost-effective, and environmentally friendly method for determining the content of multiple elements in medicinal herbs using ICP OES. The optimized experimental parameters result in low dissolved organic carbon content and residual acidity concentration. The accuracy of the method is confirmed through the analysis of certified reference materials, and its environmental friendliness is validated using the AGREE metric. Principal component analysis is used to classify the medicinal herbs based on their inorganic constituent concentrations.

Separation and characterization of related substances of Lurasidone hydrochloride by LC-QTOF-MS techniques

Unraveling the cytotoxicity and cellular uptake of low, medium and high molecular weight polyethylene glycol polymers in MCF-7 cells by green UPLC-MS/MS methods

Summary: This study investigated the cellular uptake of low, medium and high molecular weight polyethylene glycol (PEG) in MCF-7 cells using UPLC-MS/MS and CID techniques. The results showed that PEG600, PEG2K, PEG4K and PEG10K exhibited low toxicity and had limited cellular entry at concentrations of 5-1200 μg/mL. Additionally, the environmental friendliness of the developed analytical methods was evaluated.

Summary: This study employed network pharmacology-based analysis and reverse molecular docking to investigate the molecular targets and pathways associated with gynecological disorders and the potential therapeutic effects of fenugreek constituents. The results highlighted the potential of steroidal saponins, isoflavones, and flavones as active constituents of fenugreek with implications for lactation, reproductive processes, and estrogenic activity. Animal model study further supported the presence of these bioactive compounds and their effects on body weight, serum prolactin levels, serum estradiol levels, and ovarian histological structure.
